摘要
目的检测miR-421在前列腺癌(prostate cancer,PCa)患者血清和组织中的表达,并探讨其与临床病理特征的关系及意义。方法收集2015年12月~2016年12月初次在安徽医科大学第一附属医院泌尿外科住院的PCa患者血清62例及前列腺增生(prostatic hyperplasia,BPH)患者血清46例,并调取同时期病理科存档的PCa蜡块42例,BPH蜡块37例。采用qRTPCR检测血清中miR-421的表达,采用原位分子杂交法检测病理组织中miR-421的表达。结果 miR-421在PCa和BPH患者血清中的表达分别为2.52±1.70、0.82±0.65,与BPH组相比,miR-421在PCa血清中的表达升高(P<0.05),PCa患者血清和组织中miR-421的表达与Gleason评分、TNM临床分期、骨转移等有关(P<0.05),与患者年龄、术前PSA水平等均无关(P>0.05)。结论 miR-421在PCa患者血清或组织中的表达水平高于BPH患者,有望成为PCa的诊断标志物。
Purpose To detect the expression of miR-421 in serum and tissues of prostate cancer（PCa） and its clinical value in PCa.Methods 62 cases of PCa and 46 cases of benign prostatic hyperplasia（BPH） were enrolled in the Department of Urology,the First Affiliated Hospital of Anhui Medical University from December 2015 to December 2016.Another 42 cases of paraffin-embedded sections of PCa and 37 cases of BPH were also used in this study.The expression of miR-421 in serum was detected by real-time PCR（qRT-PCR）.The expression of miR-421 in tissues was detected by in situ hybridization.Results The expression of miR-421 in serum of patients with PCa and BPH was（2.52 ± 1.70） and（0.82 ± 0.65）,respectively.Compared with the expression of BPH,the expression of miR421 in serum of PCa was increased（P〈0.05）.The expression of miR-421 in serum and tissues of patients with PCa was correlated with Gleason score,TNM clinical stage,and bone metastases（P〈0.05）.It was not related to the patient＇s age,preoperative PSA level and other factors（P〉0.05）.Conclusion miR-421 is more abundant in PCa patients than that in patients with benign prostatic hyperplasia,and is expected to become a diagnostic marker for PCa.
