摘要
近年来二代测序发展迅速。目前,市场上已有多套临床自动化测序系统,突破了循环肿瘤DNA(ct DNA)在检测技术上的限制。ct DNA和循环肿瘤细胞(CTC)检测对优化结直肠癌临床治疗有重要意义。尤其是在术后复发监测及靶向药物用药后的耐药分析方面,ct DNA与CTC的应用已十分接近临床且被证明效果优于现有检验手段。此外,在结直肠癌的早期筛查领域,ct DNA也表现出巨大的应用潜力,已有实验策略可准确识别肿瘤原发灶。未来,在标准化实验流程并完善大样本临床对照研究后,液体活检将成为结直肠癌的主要检测方法。
Next generation sequencing has developed rapidly in recent years.At present,there are a number of clinical automated sequencing systems in the market,breaking through the limitation of detection technology for circulating tumor DNA(ct DNA).The detection of ct DNA and circulating tumor cell(CTC) is of great significance for optimizing the clinical treatment of colorectal cancer.Especially the application of ct DNA and CTC has been very close to the clinical practice and proved to be better than the existing test methods in the field of postoperative recurrence monitoring and drug resistance analysis after targeting drug use.In addition,in the early screening of colorectal cancer,ct DNA also has shown a great applicative potential.In the future,after standardized experimental procedures and controlled large sample clinical studies,liquid biopsy will become the main method for colorectal cancer detection.
