摘要
Runx2是一种公认的转录因子,对骨髓间充质干细胞向成骨细胞转化的过程起重要作用。体内外的研究表明,它可通过骨形态发生蛋白/Smads、丝裂原活化蛋白激酶信号通路等多种信号转导途径参与骨代谢,心血管系统的异位钙化,牙齿异常发育,肿瘤和器官纤维化等疾病的发生。另外,相关微RNA、环境因素、细胞因子、激素以及中医药提取物等对Runx2所致疾病也具有重要的调控作用。目前其在疾病的发生、发展以及预后的具体作用机制尚未完全明确,以Runx2为靶点治疗临床疾病或将成为研究热点。
Runx2 is an acknowledged transcription factor essential for osteoblast development from mesenchymal stem cells and maturation into osteocytes.Studies in vitro and in vivo have reported that Runx2 could participate bone metabolism,ectopic calcification of cardiovascular system,abnormal development of teeth,tumorigenesis,and organ fibrosis through multiple integrated complex pathways,such as bone morphogenetic protein/Smads,mitogen-activated protein kinases and others.Besides,some factors like microRNA,environment,cytokines,hormones and traditional Chinese medicine extract also play a major role in the regulation of the disease induced by Runx2.However,the function of Runx2 is not completely understood in the occurrence,development and prognosis of the diseases,and the targeting of Runx2 to treat the diseases may become a hotspot in the field.
