摘要
目的 探讨足糖萼蛋白（Podocalyxin,PODX）在非小细胞肺癌转移进程中发挥的作用及机制。 方法 使用qRT-PCR对68例非小细胞肺癌癌组织和32例癌旁组织进行PODX mRNA表达情况进行检测；继而通过WB检测人NCI-H1299细胞中PODX的表达；将设计、构建的PODX-shRNA以慢病毒包装导入NCI-H1299细胞中，同时设置shCTRL对照组，通过qRT-PCR对NCI-H1299细胞中PODX mRNA的表达进行检测，随后通过Transwell室验证PODX在不同处理组细胞迁徙其中发挥的作用，并且同时使用WB验证PODX与基质金属蛋白酶9（Matrix metalloprotein 9,MMP-9）相关。 结果 数据显示NSLCL肺癌组织中PODX mRNA表达显著高于癌旁组织（ P 〈0.01），且PODX蛋白同样存在于NCI-H1299细胞中；shPODX能够显著降低PODX的表达，且Transwell能够观察shPODX组迁徙性细胞数目显著低于未处理组及其他对照组； shPODX组细胞的胞内以及上清液中MMP-9的相对含量均低于其他3组，且与Transwell实验结果趋势一致。 结论 本研究提示PODX可能通过促进MMP-9的产生进而促进NCI-H1299细胞的迁徙，为非小细胞肺癌的分子治疗提供新的思路。
Objective： To explore the functions and mechanism of podocalyxin （PODX） in NSCLC metastasis progressions. Methods： The mRNA levels of PODX from 68 NSCLC lung tissues and 32 paracarcinoma tissues were collected and detected by RT-PCR respectively.Andthen the endogenous PODX was detectd in NCI-H1299 cells,a NSCLC cell line,by Western Blot.Subsquent the level of PODX mRNA and migration of NCI-H1299 cell with shPODX treatment and the control group shCTRLwere proved by qRT-PCR and Transwells assays.Meanwhile the realtionship between different deals for NCI-H1299 cells and MMP-9 was proved by WB. Results ： The datas showed clinical NSCLC lung tissue havehigher level mRNA of PODX than paracarcinoma tissues significantly （ P 〈0.01）.There is PODX exist in NCI-H1299 cells.When the shPODX wast ransfected into NCI-H1299 cells,the PODX mRNA have low level than untreatment cells.And the cells with shPODX decreasing number of migrating cells was detected by transwells assays.Meanwhile the the level of PODX was closely related with MMP-9 in endocellular and supernatant.The results showed the same trend with Transwells assays. Conclusion： The study suggested the PODX can promote NCI-H1299 cells migration via MMP-9.It suggests that these datas provides a novel insight about molecular therapy for lung cancer treatment.
