雷公藤红素通过活性氧诱导人卵巢癌SKOV3细胞凋亡的研究 被引量:18

Celastrol Induced Apoptosis of Human Ovarian Cancer SKOV3 Cells by Alteration of ROS Level

摘要 目的研究雷公藤红素通过细胞内活性氧(Reactive oxidative species,ROS)诱导人卵巢癌SKOV3细胞凋亡的作用机制。方法通过MTT比色法检测雷公藤红素对人卵巢癌SKOV3细胞增殖的抑制作用;通过Hoechst 33258染色荧光显微镜观察SKOV3细胞凋亡的形态学变化;通过流式细胞术检测SKOV3细胞凋亡及ROS水平;Western blot法检测凋亡相关蛋白bcl-2、cleaved caspase-3、cleaved caspase-9蛋白的表达。结果雷公藤红素对卵巢癌SKOV3细胞具有明显的增殖抑制作用,且呈时间/剂量依赖关系(P<0.05);雷公藤红素可显著增加细胞内ROS水平,诱导卵巢癌SKOV3细胞凋亡,并上调cleaved caspase-3、cleaved caspase-9蛋白的表达,下调抗凋亡蛋白bcl-2的表达(P<0.05)。结论雷公藤红素可通过提高细胞内ROS水平,诱导人卵巢癌SKOV3细胞凋亡,为卵巢癌治疗及预后方面的研究提供了新思路。 Objective To show the evidence of apoptosis in SKOV3 cells induced by celastrol via alteration of ROS level. Methods The proliferation of SKOV3 cells was determined by MTT staining method;the intracellular ROS levels were detected by flow cytometry;Hoechst 33258 staining and flow cytometry were used to detect the apoptosis of cells;and expression levels of apoptotic proteins were determined by Western Blot method. Results Celastrol had obvious proliferation inhibitory effect on ovarian cancer SKOV3 cells, and showed a time/dose dependent relationship. Celastrol induced significant apoptosis in ovarian cancer SKOV3 cells, which significantly increased intracellular ROS levels,up-regulated the levels of cleaved caspase-3 and cleaved caspase-9,and down-regulated the expression of anti-apoptotic protein bcl-2 in SKOV3 cells. Conclusion Celastrol induced apoptosis in human ovarian cancer SKOV3 cells by increasing intracellular ROS levels.
