应用离子色谱技术对胃癌患者尿液游离氨基酸进行代谢谱分析 被引量:1

摘要 目的:本实验通过检测胃癌患者和正常人尿液中游离氨基酸代谢谱,筛选出具有诊断意义的氨基酸,并探讨胃癌患者尿液氨基酸代谢变化的规律和意义。方法:留取晨尿162例分3组,采用离子色谱法检测3组尿液中22种氨基酸,应用色谱软件,构建尿液氨基酸代谢谱,对所得数据进行统计学处理。结果:胃癌组尿液氨基酸代谢谱较正常组有明显变化;不同分期胃癌组的氨基酸代谢谱也存在差异。结论:异亮氨酸等6种氨基酸可能是参与肿瘤生长和转移过程中细胞增殖和基因表达的重要代谢产物;异亮氨酸和缬氨酸在早期胃癌中可能是重要的代谢产物。 Objective: To screen out amino acids with diagnostic significance by detection of metabolic profiling of free amino acids in urine samples of both patients with gastric cancer at different stages and healthy people and to explore the regularity and significance of the changes of amino acid metabolic profiling. Methods: One hundred and sixty-two urine samples were collected and divided into 3 groups. Twenty-two amino acids in each urine sample were detected by ion chromatography(IC) and then the metabolic profiling of amino acids in the urine sample was established with Peaknet 6 software and the obtained data were statistically analyzed. Results: There existed significant differences between patients with gastric cancer and healthy people and even the patients at different stages in metabolic profiling of free amino acids in urine. Conclusion: Six amino acids including isoleucine, valine, leucine, methionine, histidine and aspartic acid may be important metabolites involved in cell proliferation and gene expression during tumor growth and metastasis and isoleucine and valine may also be important metabolites in early gastric cancer.
