摘要
目的 观察 Bcl- 2和 Bax与 PCNA在桥本甲状腺炎(HT)中表达及分布的关系 .方法 以 16例毒性甲状腺肿(NTG)为对照 ,采用 TUNEL及免疫组织化学双标记法分别检测 16例 HT患者甲状腺标本中的细胞凋亡水平及 PCNA与 Bcl- 2和 Bax的分布 .结果 凋亡率 (AR)在 HT组为(34.0± 14 .7) % ,NTG组为 (0 .9± 0 .6 ) % ;增殖率 (PR)分别为 (8.5± 3.6 ) % ,(4.2± 2 .4 ) % ;AR/PR比值各为 4 .5± 2 .4和 0 .5± 0 .9.显示 AR,PR及 AR/PR比值在 HT组均显著高于 NTG组 (P<0 .0 1) .在 HT中 ,Bcl- 2 ,Bax免疫染色阳性率分别为 88%和 82 % ;共表达细胞占 Bcl- 2或 Bax阳性细胞的比例各为 33%和 16 % .结论 HT中甲状腺滤泡细胞凋亡与增殖活动均活跃 ,凋亡水平增高更为显著 ;浸润淋巴细胞增殖活跃 ,凋亡少见 .部分甲状腺滤泡细胞 PCNA与 Bcl- 2或Bax共表达 .其凋亡和增殖的失平衡可能与调节蛋白 Bcl- 2或 Bax的作用有关 ,也可能是
AIM To investigate relationship between the expression and distribution of PCNA and those of Bcl 2 or Bax in Hashimoto's thyroiditis (HT). METHODS Specimen from nontoxic goiter (NTG, n =16) were used as the control. Apoptotic level and expressions as well as distributions of PCNA, Bcl 2 and Bax in thyroid tissues from 16 HT patients were detected by TUNEL and immunohistochemical double labelled staining repectively. RESULTS Apoptotic rates (AR) in the HT and NTG groups were (34.0±14.7)% and (0.9±0.6)% respectively, while the prolifertive rates (PR) were (8.5±3.6)% and (4.2±2.4)%, and AR/PR ratios in HT and NTG were 4.5±2.4, 0.5±0.9 respectively, which indicated that AR, PR, AR/PR ratio in HT were significantly higher than those in NTG ( P <0.01). The rates of positive immunostaining for Bcl 2 and Bax on part of thyrocytes from HT were 88% and 82%, and Bcl 2 or Bax positive percentages of co expression thyrocytes were 33% and 16%. CONCLUSION Both apoptotic and proliferative activities in thyrocytes of HT group were active, especially apoptotic level, and there was a high proliferative activity in infiltrating lymphoid cells where apoptosis is rarely seen. There was co expression of PCNA with Bcl 2 or Bax on part of thyrocytes in HT. And both Bcl 2 and Bax might regulate the imbalance between apoptosis and proliferation which may be the major mechanisms of pathologic changes in the HT group.
