解淀粉芽孢杆菌PI142产γ-聚谷氨酸发酵条件优化 被引量:3

摘要 以解淀粉芽孢杆菌PI142(Bacillus amyloliquefaciens PI142)为生产菌株,对其产γ-聚谷氨酸(γ-PGA)的发酵条件进行优化研究。以单因素实验分别考察碳源、氮源、无机盐、前体物质、初始p H值等因素对γ-PGA产量的影响。采用响应面实验设计对其发酵条件进行优化,得到PI142生产γ-PGA最佳发酵条件为:蔗糖浓度3.77%、豆饼粉浓度0.4%、谷氨酸钠浓度2.01%、磷酸氢二钾浓度0.7%、初始p H8.24,在该条件下γ-PGA产量平均值达到3.19g/L,较优化前提高了4.9倍。 Bacillus amyloliquefaciens PI142 was used as γ-Polyglutamic Acid(γ-PGA) production strain to optimize the fermentation conditions. Types and the concentration of the carbon source, nitrogen source, inorganic salts, precursor substances and initial p H value which affect the yield of γ-PGA were investigated by single factor experiment. The optimum condition for γ-PGA yielding was obtained by response surface methodology as following: 3.77% of sucrose, 0.4% of soybean cake powder, 2.01% of monosodium glutamate, 0.7% of K2 HPO4 with 8.24 of initial p H value, under which the average γ-PGA was up to 3.19 g/L and was 4.9 times higher than that of the initial fermentation.
