摘要
目的:观察肾康汤对SD大鼠慢性肾功能衰竭的影响并对其作用机制进行探讨,为其临床应用提供药理学实验依据。方法:灌胃大鼠腺嘌呤30天使其发生慢性肾衰竭,检测模型成功后给予不同浓度肾康汤和阳性药尿毒清60天,分别于第30、60天取血测肌酐(SCr)、尿素氮(BUN),治疗结束后取肾组织切片,HE染色观察其病理学变化,免疫组化检测bFGF蛋白表达情况。结果:与模型组相比,治疗组SCr、BUN明显减少,差异有统计学意义(P<0.05)。病理切片显示:空白组肾小球、肾小管形态、大小均正常,模型组肾小管极度扩张,间质可见棕黄色尿酸样结晶沉积,给药组较模型组症状明显减轻,但仍比空白组严重。免疫组化bFGF蛋白表达:阴性对照无表达,空白组微量表达,模型组大量表达,给药组表达较模型组明显减少(P<0.05)。结论:肾康汤能明显改善大鼠慢性肾衰症状,其作用机制可能与抑制bFGF蛋白表达有关。
Objective：To observe the effect of Shenkang Decoction on adenine-induced chronic renal failure in rats and to explore the mechanism,so it can provide aexperimental basis for its clinical application.Methods：Feeding these rats at the doses（2.5%）with adenine for 30 days,then giving them a cheek with different concentrations of Shenkang Decoction and positive medicine Niaoduqing for 60 days after the model being successed,and measuring Serum creatinine（SCr）and urea nitrogen（BUN）at the thirty days and sixty days,observing the pathological changes of kidney by HE staining and detecting the expression of basic fibroblast growth factor（bFGF）by immunohistochemistry.Results：Compared with the model group,SCr and BUN of the groups,which treated with protection drugs,were significantly reduced,and the difference was statistically significant（P〈0.05）.Pathology showed：in blank group,the morphology of glomerular and tubular are normal,but in model group,renal tubular extreme expansion,more interstitial inflammatory cell infiltration and brown color urate crystal deposition,however,it significantly reduced after treatment.As the immunohistochemical shown,there were no expression of bFGF in negative control and few in the blank group,while a lots of bFGF can be founded in the model group,but significantly reversed by medicine（P〈0.05）.Conclusion：Shenkang Decoction have definitely therapeutic effect on CRF,which may be related to inhibit the expression of bFGF.
