摘要
目的建立反相高效液相色谱法(high performance liquid chromatography,HPLC)测定保健食品中功效成分腺苷。方法样品中腺苷经85%的甲醇溶液加热回流提取,C18反相柱分离,在波长260 nm处进样检测。结果在5.045~252.250μg/m L范围内具有良好的线性关系,回归方程是Y=27281X-57639,相关系数r=0.9999,检出限为0.5μg/m L(S/N=3),此方法的回收率为91.5%~97.7%,相对标准偏差(relative standard deviation,RSD)为1.5%~2.8%。结论此方法简便快速,准确可靠,可用于各类保健食品中腺苷含量的测定。
Objective To establish a method for determination of adenosine of functional ingredients in health food by reversed phase-high performance liquid chromatography(HPLC). Method The adenosine from sample was heating refluxed by 85% methanol, and then separated by C18 column and detected at the wavelength of 260 nm. Results There was a good linear relationship in the range of 5.045-252.250 μg/m L. The regression equation was Y=27281 X-57639, with the correlation coefficient(r) of 0.9999, and the limit of detection was 0.5 μg/m L(S/N=3). The recoveries were 91.5%-97.7%, and relative standard deviations(RSDs) were 1.5%-2.8%. Conclusion This method is simple, rapid, accurate and reliable, which can be used for the determination of adenosine content in all kinds of health food.
