摘要
目的探究表皮生长因子受体Ⅲ型突变体(EGFRvⅢ)及血小板衍生生长因子-D(PDGF-D)在结肠癌组织中的表达及对靶向治疗的指导作用。方法选取2016年1月2017年6月于我院行结肠癌根治术的患者60例,收集其标本作为观察组,另取正常结肠组织20例作为对照组。采用免疫组织化学技术及蛋白质印迹法检测组织中的EGFRvⅢ和PDGF-D,比较结肠癌和正常结肠组织中上述2指标的表达差异及影响表达的因素。结果观察组EGFRvⅢ和PDGF-D的阳性表达率及含量均高于对照组,差异均有统计学意义(P<0.05)。EGFRvⅢ和PDGF-D在结肠癌中的表达与性别和年龄无关,而同分化程度和临床病理分期(TNM)有关,分化程度低、TNM分期高的标本EGFRvⅢ和PDGF-D阳性表达程度及含量均高于分化程度高、TNM分期低的标本,差异均有统计学意义(P<0.05)。EGFRvⅢ和PDGF-D在结肠癌组织中的表达呈正相关(r=0.647,P<0.05)。结论 EGFRvⅢ和PDGF-D在结肠癌组织中表达显著增加,且同分化程度和TNM分期有关。
Objective To investigate the expressions of epidermal growth factor receptor variant type Ⅲ(EGFRvⅢ) and platelet derived growth factor-D(PDGF-D) in colorectal carcinoma tissues and their guiding role for targeted therapy. Methods Sixty patients, who were treated with radical resection of colon cancer in our hospital between January 2016 and June 2017, were included in the study, and their specimens were collected as the study group. Another 20 cases of normal colon tissue were included as the control group. Immunohistochemistry and Western Blotting were used to examine the expressions of EGFRvⅢ and PDGF-D in the tissues. The differences in the two indexes and the influencing factors of the expressions between the colon cancer and normal colon tissues were compared. Results According to the immunohistochemistry and Western Bloting, the positive expression rates and levels of EGFRvⅢ and PDGF-D in the study group were significantly higher than those in the control group, and the differences were statistically significant(P〈0.05). The expressions of EGFRvⅢ and PDGF-D were not related to gender and age, but were related to the degree of differentiation and the clinical tumor node metastasis(TNM). The positive rates and levels of EGFRvⅢ and PDGF-D in the specimens with low differentiation and high TNM stage were higher than those in the specimens with high differentiation and low TNM stage, and the differences were statistically significant(P〈0.05). The expressions of EGFRvⅢ and PDGF-D in the colon cancer tissues were positively correlated(r=0.647,P 〈0.05). Conclusion The expressions of EGFRv Ⅲ and PDGF-D in colon cancer tissues significantly increase, which are correlated with the degree of differentiation and TNM stage.
