肺机械扩张增进表面活性物质分泌机制的研究

摘要 采用大鼠离体肺灌流模型,观察肺泡灌洗液中肺表面活性物质(PS)含量的变化。结果表明:在增大潮气量机械扩张肺时PS分泌显著增加(p<0.05),该分泌增加现象能被细胞松弛素B及硝苯吡啶所抑制(p<0.05),但不为细胞外缺钙所抑制(p>0.05)。从而提示扩张所引起的PS分泌增加可能与细胞内的收缩结构有关,而与细胞外钙离子无关。 In order to study the mechanism of pulmonary surfactant secretion stimulated by mechanical expansion of lung, an isolated perfused lung (IPL) model ofWistar rat (200-300g) was established. The modified Kreb-bicarbonate perfusate was used. In condition of excluding the influences of nerves and hormones, the amount of surfactant in lung lavage fluid was detected after lung was expanded.The surfactant in lavage fluid was extracted by the method of Bligh and Dyer following being digested with HClO4. Phosphorus was detected with colorimetric analysis. It was observed that in expansion of the lung by increase the tidal volume the surfactant content increased significantly in the lavage fluid (p<0.05). This phenomenon of increase could not be inhibited by calcium depletion in the perfusate. but could be inhibited by cytochalasin B and nifedipine (p<0.05). These results suggest that the intracellular contractile structure plays an important role in the surfactant secretion stimulated by mechanical expansion of the lung.
