伏马菌素B1对BALB/c小鼠的亚急性经口毒性研究

摘要 目的研究伏马菌素B1(FB1)对BALB/c小鼠的亚急性经口毒性。方法将30只健康SPF级BALB/c小鼠按体重随机分为三组,分别为对照(去离子灭菌水)组和0.75、7.50 mg/kg FB1染毒组,每组10只,雌雄各半。采用经口灌胃方式进行染毒,染毒容量约为0.04 ml/g,每天1次,连续染毒28 d。观察小鼠的一般情况,检测血液学指标(全血白细胞计数、红细胞计数、血红蛋白含量、红细胞比积、血小板计数、白细胞分类),生化学指标[血清(丙氨酸氨基转换酶(ALT)、天门冬氨酸氨基转换酶(AST)、总蛋白(TP)、白蛋白(ALB)、尿素氮(BUN)、肌酐(CRE))],观察主要脏器的病理学改变并计算脏器系数。结果观察期内,小鼠未出现中毒和死亡现象。各组小鼠未出现体重、进食量和食物利用率的变化。各组小鼠的肝脏、脾脏、肾脏、睾丸和子宫系数间比较,差异均无统计学意义(P>0.05)。与对照组比较,7.50 mg/kg FB1染毒组雌性小鼠的血清ALT和AST活力均升高(P<0.05)。对于雄性动物,7.50 mg/kg FB1染毒组的白细胞计数降低(P<0.05),红细胞计数升高(P<0.05);而对于雌性动物,0.75 mg/kg FB1染毒组的中性粒细胞百分比降低和淋巴细胞百分比上升,7.50 mg/kg FB1染毒组的中性粒细胞降低及单核细胞百分比升高。另外,7.50 mg/kg FB1染毒组小鼠出现肝脏病理学改变,其余脏器病理学组织观察未见异常。结论伏马菌素B1对小鼠具有肝毒性,且雌性小鼠比雄性小鼠更为敏感。 Objective To test the subacute oral toxicity of purified fumonisin B1(FB1) to BALB/c mice. Methods A total of 30 SPF level BALB/c mice were randomly divided into three groups,control group(sterile deionized water) and two FB1-treated groups(0.75 and 7.50 mg/kg body weight),10 in each, half female and half male.The treatment was conducted through gavage once a day at 0.04 ml/g, for 28 consecutive days. The hematology and clinical chemistry biomarkers were detected.Histopathological examination was carried out and the organ coefficients were calculated. Results No poisoning and death were observed in all the rats during the study. By the end of the experiment,no significant changes were seen in body weight,food consumption and organ coefficients among groups. Compared with the control group, in high dose group,activities of ALT and AST were increased in females, WBC counts decreased and RBC counts increased significantly in males, and lymphocytes increased and neutrophils decreased significantly in females(P〈0.05). Significant pathological changes in livers were observed for males and females in 7.50 mg/kg body weight group. Conclusion Subacute oral toxicity study indicates that fumonisin B1 has obvious hepatotoxicity to mice, especially to females.
