新疆黑蜂胶对结肠癌大鼠模型中调节性T细胞的影响 被引量:3

Effects of Xinjiang black propolis on regulatory T cells in colon cancer rat model

摘要 目的探讨新疆黑蜂胶调控结肠癌大鼠模型中调节性T细胞(Tregs)功能进而对结肠癌治疗作用的研究。方法雄性Wistar大鼠随机分组,空白组只进行正常喂养,模型组给予N-甲基-N′-硝基-N-亚硝基胍(MNNG)灌肠12周,蜂胶预防组于灌肠期间予以黑蜂胶进行预防,溶剂对照组给予33%乙醇灌肠,模型复制成功后将模型组随机分为阴性对照组(建模但不给予治疗),蜂胶治疗组(给予黑蜂胶治疗30 d)以及阳性对照组(给予5-FU治疗30 d)。HE染色观察结肠癌变组织学变化,采用流式细胞术、RT-PCR法、ELISA法、免疫组化方法检测Tregs细胞表型及与其增殖分化相关因子IL-10、TGF-β的表达。结果阴性对照组大鼠体质量、血清中TGF-β较空白组相比显著降低(P<0.05);治疗组大鼠血清中TGF-β的水平显著高于阴性对照组(P<0.05);与空白组相比,阴性对照组大鼠结肠大体及病理切片均有明显变化;阴性对照组大鼠血清中IL-10及脾脏中CD4+CD25+Treg细胞亚群比例较空白组显著增高(P<0.05);蜂胶治疗组和阳性对照组大鼠血清中IL-10及CD4+CD25+Treg细胞亚群的比例低于阴性对照组,差异有统计学意义(P<0.05)。结论新疆黑蜂胶可通过调节Tregs的免疫抑制功能及相关因子的表达发挥抗肿瘤作用,进而对结肠癌的治疗以及预后产生影响。 This study aimed to investigate the effect of Xinjiang black propolis on the regulatory T cells (Tregs) in colon cancer model and its therapeutic effect on colon cancer. Male Wistar rats were divided randomly into 4 groups: blank group (only fed with normal feeding), model group (given N-methyl-N'-nitro-N-nitrosoguanidine (MNNG) enema for 12 weeks), prevention group (treated with black propolis during the enema), and solvent control group (given 33% ethanol enema). After the colorectal cancer model replication was constructed, the model rats were randomly divided into negative control group (only model replication without drug treatment), propolis treatment group (treatment of 30 d with black propolis), was used to observe the histological changes pos of itive control group (treatment of 30 d with 5-FU). Then HE staining colon cancer. The phenotype of Tregs cells and the expression of proliferation- and differentiation- related factors IL-10 and TGF-13 were detected by flow cytometry, RT-PCR, ELISA and immunohistochemistry. Data showed that the body weight and the TGF-β in the serum of the negative control group were significantly lower than those in the blank group (P〈0.05); the serum TGF-β level of the rats in the treatment group was significantly higher than that in the negative control group (P〈0.05); Compared with the blank group, the colonic and pathological sections of the rats in the negative control group were obviously changed; the level of serum IL-10 and the proportion of CD4^+CD25^+Treg cell subsets in the spleen of the negative control propolis treatment group and positive control group were significantly lower than those in the negative control group (P〈0.05). Taken together, Xinjiang black propolis plays function of Tregs and the expression of related factors, then an antitumor role by regulating the immunosuppressive influences the treatment and prognosis of colon cancer
