摘要
本研究旨在探讨基因单核苷酸多态性(single nucleotide polymorphisms)与传统危险因素相互作用对苗族高血压发病的影响。选取212例苗族人群作为研究对象,对6个SNP位点(ALDH2-rs671,ATP2B1(rs17249754,rs2681472),CASZ1-rs12046278,MTHFR-rs17367504,ZNF652-rs16948048)进行Snapshot基因分型,并应用逻辑回归分析基因与环境的交互作用。研究结果显示:(1)6个SNPs的基因型分布均符合Hardy-Wenberg遗传平衡定律(p〉0.05),在高血压实验组与对照组的比较中,6个SNP位点多态性基因频率分布差异无统计学意义;(2)二元逻辑回归分析显示BMI、TG、LDL、血脂异常和年龄可能为苗族高血压患病的危险因素;(3)将潜在危险因素带入逻辑回归模型中,分析发现肥胖与ZNF652-rs16948048、MTHFR-rs173-67504基因位点多态性的交互作用与苗族EH发病存在关联性。综上所述,传统危险因素-肥胖与ZNF652-rs16948048、MTHFR-rs17367504基因多态性的交互作用可能对苗族高血压的发生有一定影响。
This study was to explore the influence that Miao minority's hypertension morbidity was affected by the interaction between gene single nucleotide polymorphism and traditional risk factors.Choosing 212 Miao people as the research object,the Snapshot genotyping was used in six loci(ALDH2-rs671,ATP2B1(rs17249754,rs26814-72),CASZ1-rs12046278,MTHFR-rs17367504,ZNF652-rs16948048)and the interaction between gene and environment w as analyzed by logistic regression.The results showed that the distribution of six SNPs genotypes accorded with Hardy-Wenberg law of genetic equilibrium(p〉0.05).The distribution of six SNP loci polymorphic gene frequency was not statistically significant in the comparison of the case group and control group.The binary logistic regression analysis showed that BMI,TG,LDL,dyslipidemia and age would be risk factors for hypertension of Miao people.Bringing the potential risk factors into the logistic regression model,the analysis was found that the interaction which was between obesity and ZNF652-rs16948048、MTHFR-rs17367504 gene loci polymorphism,was linked with the EH morbidity of Miao people.In conclusion,the interaction which was between obesity,the traditional risk factor,and ZNF652-rs16948048、MTHFR-rs17367504 gene polymorphism,would affect the development of hypertension of Miao People.
