摘要
目的 制备分泌抗 HBs Ag单克隆抗体杂交瘤细胞株 ,为建立快速诊断 HBs Ag的方法奠定了基础。 方法 通过细胞融合建立了能稳定分泌抗 HBs Ag单抗的杂交瘤细胞株 ,利用 EL ISA技术测定单抗的特异性 ,并用此单抗对临床标本进行初步检测。 结果 细胞株分泌的单抗可与 HBs Ag特异性结合 ,与乙肝病毒 E抗原和核心抗原无交叉反应 ,经免疫扩散试验分析为 Ig G1型 ,将此单抗用辣根过氧化物酶标记后可与 HBs Ag阳性的病人血清反应。 结论 筛选出了一株适合临床应用的单抗杂交瘤细胞株。
Objective To construct anti HBsAg hybridoma cell line,this will be helpful to the establishment of rapid HBsAg diagnosis assay. Methods A hybridoma cell line which consistently secreted monoclonal antibody (McAb) against HBsAg was obtained through cell fusion.The specificity of the McAb was analysed by ELISA. The McAb was used for detecting some clinical samples. Results ELISA analysis showed that the McAb specifically reacted to HBsAg, no cross reaction to HBeAg and HBcAg. The sub class of the McAb was determined to be IgG1 by immune diffusion test .Labled with horeseradish peroxidase (HRP), the McAb can react with HBsAg in serum samples from HB patient. Conclusion The hybridoma cell line may be applicable to the diagnosis of HBsAg.
