摘要
目的:比较正常及炎症来源的牙周膜干细胞(periodontal ligament stem cells,PDLSCs)中线粒体融合蛋白-1(mitofusion 1,Mfn1)的表达差异及其对PDLSCs成骨分化的影响。方法:分离培养PDLSCs,分别为正常来源(HPDLSCs)、牙周炎来源(P-PDLSCs)及应用IL-1β模拟炎症微环境诱导牙周膜干细胞(H-PDLSCs+IL-1β)。RT-PCR方法检测对比H-PDLSCs与P-PDLSCs、H-PDLSCs与H-PDLSCs+IL-1β中Mfn1表达差异;RT-PCR检测成骨基因、茜素红染色及氯化十六烷基吡啶定量,对比P-PDLSCs及si Mfn1下调Mfn1表达后的成骨分化能力。结果:与H-PDSLCs组相比,P-PDSLCs组及H-PDLSCs+IL-1β组中Mfn1表达量显著升高(P<0.05);si Mfn1下调P-PDSLCs中Mfn1表达量后,其成骨分化能力增强(P<0.05)。结论:炎症微环境下,PDLSCs中Mfn1表达量升高,导致成骨分化能力下降。
Objective: To detect the expression of mitofusion-1 (Mfnl) in periodontal ligament stem cells (PDLSCs) isolated from healthy and periodontitis tissue and to study the effect of Mfnl on the osteogenic differentiation of PDLSCs. Methods: PDLSCs were isolated from the healthy and periodontitis human samples (H-PDLSCs and P-PDLSCs). IL-1β was applied to mimic the inflammation microenvironment( H-PDLSCs + IL-1 β ). RT-PCR was used to detect the expression of Mfnl in H- PDLSCs, P-PDLSCs and H-PDLSCs + IL-1β. The expression of Mfnl in P-PDLSCs was down-regulated by siRNA of Mfnl (siMfnl). The osteogenic differentiation of the cells was examined by RT-PCR, alizarin red staining and cetyl pyridine chlo- ride quantitative analysis. Results: The expression level of Mfnl in P-PDLSCs and H-PDLSCs + IL-1β (5 ptg/ml) groups was higher than that in H-PDLSCs(P 〈0.05). When the expression of Mfnl in P-PDLSCs was down-regulated by siMfnl the os- teogenic differentiation ability of P-PDSLCs was restored( P 〈 0.05 ). Conclusion: Inflammation may promote Mfnl expression in PDESCs and inhibite the osteogenic differentiation of P-PDLSCs.
