摘要
目的:在体外培养条件下,研究表观遗传相关的组蛋白去乙酰化酶抑制剂丙戊酸钠(VPA)对依赖利福平(R)结核分支杆菌菌株生长的影响。方法:采用匡氏琼脂培养基及技术,分别观察临床分离R依赖株和R耐药株在含R和VPA的培养基中生长情况,探究VPA对结核菌利福平依赖性的影响。结果:利福平依赖株在含R+VPA的培养基上生长较在仅含R培养基上生长落数和旺盛程度均下降;而利福平耐药株在含R+VPA的培养基上生长与在仅含R培养基上生长落数和旺盛程度比较,未见明显差别。结论:VPA在体外可显著抑制结核分支杆菌对利福平的依赖性。
Objective: To investigate the effect of epigenetic-related histone deacetylase inhibitors( HDACIs) VPA on the growth of rifampin-depending mycobacterium tuberculosis strains on cultural medium. Methods: The isolation and the culture of the colonies were done by Kuang' s agar medium and technique to investigate the effect of VPA on the characteristics of rifampin depending mycobacterium tuberculosis. The growth characteristics of clinical isolated strains of rifampin dependent tuberculosis and strains of rifampin resistant tuberculosis on cultural medium containing rifampin and on cultural medium containing VPA were observed and compared respectively. Results: Colonies of clinical isolated rifampin dependent tuberculosis grew faster and more vigorous on cultural medium containing rifampin than that on cultural medium containing VPA and rifampin,and no significant difference was found between the growth of clinical isolated strains of rifampin resistant tuberculosis on cultural medium containing VPA and rifampin and on cultural medium containing rifampin. Conclusion: VPA can significantly inhibit the rifampin dependence feature of mycobacterium tuberculosis.
