摘要
目的探讨Aurora激酶A(Aurora-A)、Aurora激酶B(Aurora-B)与宫颈癌发生发展的相关性及意义。方法回顾性选取宫颈癌标本(鳞状细胞癌)30例为宫颈癌组;宫颈上皮内瘤样病变(CIN)30例为CIN组;正常宫颈组织30例为正常组。采用免疫组化检测Aurora-A、Aurora-B、采用半定量RT-PCR法检测高危人乳头状瘤病毒(HPV16及HPV18)的mRNA在不同宫颈组织的表达,结合宫颈癌的病理特征进行分析。结果 (1)Aurora-A、Aurora-B及HPV16 mRNA、HPV18 mRNA在正常组、CIN组及宫颈癌组的表达分别为:1.54%、2.09%、(41.17±8.02)%、(16.05±6.78)%;36.58%、29.32%、(64.52±14.21)%、(28.97±7.98)%及71.59%、68.55%、(91.18±21.45)%、(46.62±13.58)%,宫颈癌组高于CIN组及正常组,差异有统计学意义(P<0.05),CIN组高于正常组,差异有统计学意义(P<0.05)。Aurora-A、Aurora-B及HPV16 mRNA、HPV18 mRNA与宫颈病变呈正相关(P<0.05),Aurora-A或AuroraB均与HPV16 mRNA、HPV18 mRNA的表达呈正相关(P<0.05)。(2)Aurora-A或Aurora-B均与TNM分期、淋巴结转移、组织学分级呈正相关(P<0.05)。结论 Aurora-A、Aurora-B的表达与高危HPV有密切的关系,随着子宫颈病变的进展而逐渐增加,Aurora-A与Aurora-B有望成为宫颈癌早期诊断的分子标记以及的预防、治疗宫颈癌的新靶点。
Objective To study the correlation between the Aurora A( Aurora-A) and Aurora B( Aurora-B) protein kinases and the occurrence and development of cervical cancer. Methods Cervical carcinoma tissues( squamous carcinoma) from 30 patients were selected as cervical carcinoma group,30 cases of cervical intraepithelial neoplasia( CIN) tissues were selected as CIN group,and 30 cases of normal cervical tissues were selected as normal group. Aurora-A and Aurora-B were detected by immunohistochemical method. The expression of mRNA of high-risk human papilloma virus( HPV16 and HPV18) in different cervical tissues were detected by semi-quantitative RT-PCR technology. And their relationships with clinical pathological features were analyzed. Results(1)The expression level of Aurora-A,Aurora-B,HPV16 mRNA and HPV18 mRNA were 1. 54%,2. 09%,( 41. 17 ± 8. 02) %,( 16. 05 ± 6. 78) %; 36. 58%,29. 32%,( 64. 52 ± 14. 21) %,( 28. 97 ± 7. 98) %and 71. 59%,68. 55%,( 91. 18 ± 21. 45) %,( 46. 62 ± 13. 58) % in normal group,CIN group and cervical carcinoma group respectively. The expression level of those in cervical cancer group were statistically higher than those in CIN group( P 〈 0. 05). The expression level of those in CIN group were statistically higher than those in normal group( P 〈 0. 05). There was positive correlation between Aurora-A,Aurora-B,HPV16 mRNA or HPV18 mRNA and the degrees of cervical lesions. There was positive correlation between the expression level of Aurora-A or Aurora-B and HPV16 mRNA or HPV18 mRNA( P 〈 0. 05).(2)There were positive correlation between Aurora-A or Aurora-B and the TNM staging,lymph node metastasis or histological grading-scale( P 〈 0. 05). Conclusion The expression of Aurora-A and Aurora-B were closely related to high-risk human papilloma virus,with the progress of the malignant diseaseand,Aurora-A and Aurora-B also increase. Aurora-A and Aurora-B May be molecular markers in early diagnosis of cervical cancer and novel targets for prevention and treatment of cervical cancer.
