摘要
目的利用高效液相色谱-蒸发光散射法(HPLC-ELSD)测定14批次黑参中4种稀有皂苷20(S)-Rg3、20(R)-Rg3、Rk1、Rg5的含量。方法采用HPLC-ELSD对黑参中4种稀有皂苷进行测定。色谱条件:采用COSMOSIL C18-PAQ色谱柱(4.6 mm×250 mm,5μm),以80%乙腈水溶液(A)-10%乙腈水溶液(C)为流动相进行梯度洗脱,流速1.2 m L·min-1,柱温30℃,漂移管温度为60℃,进样体积20μL。结果稀有皂苷20(S)-Rg3、20(R)-Rg3、Rk1、Rg5得到良好的分离,分别在21~620、19~570、20~610和19~560μg·m L-1与色谱峰面积呈良好的线性关系。14个黑参样品中稀有皂苷20(S)-Rg3的含量范围为0.107%~0.233%,20(R)-Rg3的含量范围为0.075%~0.220%,Rk1的含量范围为0.161%~0.330%,Rg5的含量范围为0.442%~0.822%。结论 HPLC-ELSD法可用于同时测定黑参中4种稀有皂苷20(S)-Rg3、20(R)-Rg3、Rk1、Rg5成分,且方法简单、可靠、易行。同时,其结果为以后建立和完善黑参的炮制工艺及质量标准提供一定的参考依据。
Objective To determine the content of 4 rare ginsenoside 20 (S)-Rg3, 20 (R)-Rg3, Rkl, and Rg5 in 14 batches of black ginseng by HPLC-ELSD. Methods HPLC-ELSD was used to determinate 4 rare ginsenosides in black ginseng. The chromatographic separation was performed on a COSMOSIL C18-PAQ (4.6 mm×250 mm,5μm). The gradient elution was carried out in 80% acetonitrile aqueous solution (A)-10% acetonitrile aqueous solution (C) as the mobile phase. The flow rate was 1.2 mL min-i, with the column maintained at 30 ℃. ELSD was set to a probe temperature at 60 ℃. The injected sample volume was 20 μL. Results Good lilearity of the 4 rare saponins was shown between 21 -- 620 μg mL-1, and 19 -- 570 μg mL-1, 20 -- 610 μg mL-1, 19 -- 560 μg mL-1 for 20 (S)-Rg3, 20 (R)-Rg3, RK1 and Rgs, respectively. The content of rare ginsenoside 20 (S)-Rg3 in the samples of 14 black ginseng was 0.107% -- 0.233%, that 20 (R)-Rg3 was 0.075% -- 0.220%, that Rk was 0.161% -- 0.330%, and that Rg5 was 0.442% -- 0.822%. Conclusion HPLC-ELSD method can be used to simultaneously determinate 4 rare ginsenoside 20 (S)-Rg3, 20 (R)-Rg3, Rkl and Rgs. The method is simple, reliable and easy to operate. The results provide a reference basis for the establishment and improvement of the processing technology and quality standards of black ginseng.
