不同剂量HepG2细胞诱导小鼠胸腺产生差异性免疫应答

摘要 目的:探讨小鼠胸腺对不同剂量HepG2细胞免疫效应的差异。方法:将不同剂量DiI标记的HepG2细胞(2×10~3个/μL、2×10~5个/μL、2×10~7个/μL)注射到小鼠胸腺内,检测不同时间检点(3d,5d,7d,14d,21d,28d)小鼠胸腺出现的免疫应答变化。结果:低剂量HepG2细胞在小鼠胸腺内没有形成肿瘤组织,7d被清除;中、高剂量HepG2细胞在小鼠胸腺内均形成肿瘤组织,其周围出现不同程度T淋巴细胞聚集,并在不同时间点(中剂量14d,高剂量21d)被完全清除。结论:胸腺可对异种抗原产生免疫应答,其应答程度与抗原剂量有关。 Objective:To investigate the difference immune effects produced by mice thymuses after intrathymic injection of different doses of human HepG2 cells.Methods:Different doses of DiI-labeled HepG2 cells(2×10~3个/μL、2×10~5个/μL、2×10~7个/μL)were injected into the thymuses of mice,then immune responses in the thymuses of mice examined at different times(3 d,5 d,7 d,14 d,21 d,28 d).Results:Low-dose HepG2 cells did not form tumor tissue in the thymuses of mice and were cleared on 7 th day;Medium and high doses of HepG2 cells formed tumor tissue in the thymuses of mice,with varying degrees of T lymphocyte aggregation around them,and were completely removed at different time points(medium dose 14 d,high dose 21 d).Conclusion:Thymus can produce immune response to heterologous antigens,and the degree of immune response relate to the dose of antigen.
