绵羊mtDNA COⅠ基因作为DNA条形码鉴定品种及系统进化可行性分析 被引量:4

摘要 以山东小尾寒羊、洼地绵羊、苏尼特羊、河北小尾寒羊和湖羊为研究对象,采用测序方法获得线粒体DNA(mtDNA)COⅠ基因序列,利用COⅠ基因片段(58~705bp)进行群体遗传学和遗传多样性分析,探讨mtDNACOⅠ基因作为DNA条形码基因鉴定不同绵羊品种和系统进化的可行性。结果表明:不同绵羊种群间相似性较高(99.1%~100.0%),就种群的单倍型及分布特点而言,河北小尾寒羊7只个体有独立单倍型,但每个单倍型仅有1只个体,未发现某个种群的特有单倍型,说明COⅠ基因(58~705bp)不适于上述种群的鉴别。系统进化树分析结果表明,河北小尾寒羊与洼地绵羊先聚在一起,然后与湖羊聚在一起,再与苏尼特羊、滩羊、山东小尾寒羊聚在一起,最后与阿勒泰羊聚在一起,该结果与这些种群的历史迁徙、种群演化一致。COⅠ基因片段(58~705bp)可用于绵羊系统进化分析,但不适于作为DNA条形码序列用于不同绵羊种群鉴定,有待于扩大品种数量和群体数量进一步验证。 The mtDNA CO I gene, of which Shandong Small-tailed Han sheep, Wadi sheep, Sunite sheep, Hebei Small-tailed Han sheep and Hu sheep, were obtained through mitochondrial genome sequencing. The population genetics and genetic diversity were analyzed by CO I gene segment (58--705 bp), as well as the feasibility of mtDNA CO I gene as barcoding gene was explored in ovine breed identification and phylogenetic in this experiment. The result indicated that the comparability of different sheep population was 99.1%--100.0%. From the haplotypes and distributions of popula- tions, there were independent haplotypes which contained one individual in seven samples of Hebei Small-tailed Han sheep, however, the flock specific haplotype was no found. The above result demonstrated that CO I gene (58--705 bp) was not suitable for population identification. The phylogenetic analysis showed that the Hebei Small-tailed Han sheep and Wadi sheep were clustered first, and then they clustered with Hu sheep. Further, they clustered with Shandong Small-tailed Han sheep, Tan sheep and Sunite sheep. Ultimately, Altay sheep clustered with the above populations. The results were consistent with history migration and population evolution. The CO I gene segment (58--705 bp) could be used to phylogenetic analysis. However, it was not suitable for DNA barcoding which used to identify different sheep populations. This conclusion was further confirmed through more quantities and populations.
