发酵罐法培养疣孢漆斑菌产漆酶培养条件的研究 被引量:1

摘要 提高疣孢漆斑菌的漆酶产量,并且降低成本,使其得到大规模的应用,还要依赖于成熟的工业化发酵技术,因此实验选用16L全自动不锈钢发酵罐对疣孢漆斑菌菌种进行发酵培养,主要通过对不同通气量、搅拌速率和补加营养物质来研究疣孢漆斑菌发酵罐培养产漆酶的培养条件,以提高菌种漆酶产量。结果表明,通气量为1.0vvm时适于疣孢漆斑菌产漆酶,并且不会对设备产生过高的损耗;搅拌器转速为150r/min时,可以有效提高发酵液溶氧量,控制菌种生长,提高产酶活力;补加营养物质的研究显示,虽然没有达到预期的使产酶量出现二次高峰的效果,但补加营养物质可以延长菌种生长时间。 Mature industrial fermentation technology can increase the production of laccase prodused by myroth- ecium verrucaria and reduce cost, which makes it be used widely. In this paper, myrothecium verrucaria was cul- tured to produce laccase. The effects of different ventilatory capacities, stirring speeds and nutrient supplement on production of laccase from myrothecium verrucaria were studied to increase the production of laccase. The result showed that 1.0vvm of ventilatory capacity was suitable for myrothecium verrucaria to produce laccase and could not damage equipments~ 150r/min of stirring speed could enhance dissolved oxygen in culture medium and control the growth of strains and therefore enhanced the production of laccase; the study about the effect of nutrient supple- ment on laccase production showed that the supplement of nutrient did not achieve the expected results to enhance the production of laccase, but could lengthen the growth of strains.
