摘要
目的研究藻蓝蛋白对家兔心肌缺血-再灌注损伤模型的影响。方法家兔60只,采用随机数字表编号后随机分为假手术组,缺血-再灌注组,藻蓝蛋白1、2、3组,除假手术组外均建立心肌缺血-再灌注损伤模型。成模后2 h,藻蓝蛋白1、2、3组用藻蓝蛋白混悬液50、200、400 mg/(kg·d)灌胃2周,假手术组和缺血-再灌注组灌等体积生理盐水对照。采用酶联免疫吸附测定法检测2周时家兔各组大鼠血清中肌酸激酶同工酶MB(CK-MB)、一氧化氮(NO)、超氧化物歧化酶(SOD)活性、内皮素-1(ET-1)、内皮型一氧化氮合酶(e NOS)及心肌组织丙二醛(MDA);免疫组化染色检测诱导型一氧化氮合酶(i NOS)阳性表达;反转录聚合酶链反应(RT-PCR)检测缺血区血管内皮生长因子(VEGF)m RNA的表达。结果藻蓝蛋白2、3组NO和SOD、e NOS明显提高,i NOS阳性细胞数增多,缺血区组织VEGF、VEGFm RNA高表达,ET-1、CK-MB和心肌MDA显著降低,与模型组比较(P<0.05),差异有统计学意义。结论藻蓝蛋白对家兔心肌缺血-再灌注损伤有保护作用。
Objective To investigate the effect of phycocyanin on myocardial ischemia-reperfusion injury in rabbits. Methods 60 rabbits were randomly numbered and divided into sham operation group, ischemia reperfusion group and phycocyanin groups1,2,3, myocardial ischemia-reperfusion injury models were established in rabbits except the sham operation group. After modeling for 2 h, the three phycocyanin groups were treated by different dose of phycocyanin suspension of 50, 200, 400 mg/(kg·d) by gavage for 2 weeks; the sham operation group and ischemia reperfusion group were given the same volume of saline as control. The ELISA assay was used to test the level of MB isoenzyme of CK(CK-MB), nitric oxide(NO), superoxide dismutase(SOD) activity, endothelin-1(ET-1), endothelial nitric oxide synthase(eNOS) and malondialdehyde(MDA) in rabbit serum; the positive expression of inducible nitric oxide synthase(iNOS) was detected by the immunohistochemical staining; reverse transcription polymerase chain reaction(RT-PCR) was used to detect the mRNA expression of vascular endothelial growth factor(VEGF) in ischemic regions. Results In the phycocyanin group 2 and group 3, the expression level of NO, SOD and eNOS significantly increased, and the number of iNOS positive cells also increased; in the ischemic tissues, the high expressions of VEGF can be detected, but the expression level of ET-1, CK-MB and MDA significantly decreased compared with the modelgroup(P〈0.05), the difference was statistically significant. Conclusion The phycocyanin has protective effects on myocardial ischemia reperfusion injury in rabbits.
