摘要
采用浓硫酸法对白背三七多糖(Gynura davaricata(L.)DC polysaccharides,GDPs)进行硫酸酯化修饰,得到取代度为1.23的硫酸酯化多糖(sulfated GDPs,S-GDPs)。进一步分析GDPs和S-GDPs清除1,1-二苯基-2-三硝基苯肼自由基(1,1-diphenyl-2-picrylhydrazyl,DPPH)、超氧阴离子自由基(O2-·)、羟自由基(·OH)的能力,并考察两种多糖对H_2O_2诱导的红细胞氧化损伤以及·OH诱导的小鼠肝脏线粒体氧化损伤的保护作用。研究表明:GDPs和S-GDPs均具有抗氧化活性,其中S-GDPs清除DPPH(P<0.01)、O2-·(P<0.05)以及·OH(P<0.05)的能力显著高于GDPs;S-GDPs能显著降低红细胞溶血度(P<0.01)及其丙二醛含量(P<0.05),显著降低小鼠肝脏线粒体丙二醛含量(P<0.05)、极显著增加线粒体细胞膜的琥珀酸脱氢酶(SDH)和ATP酶(Na+-K+-ATPase、Ca^(2+)-Mg^(2+)-ATPase)活性(P<0.01),作用优于GDPs。由此可见,硫酸酯化修饰可以增强GDPs的抗氧化活性。
Sulfuric acid was used in the sulfating polysaccharides from Gynura davaricata(L.)DC.(GDPs),and substituted degree of sulfated polysaccharides(S-GDPs)of 1.23 was obtained.Then,the scavenging capacities of GDPs and S-GDPs against 1,1-diphenyl-2-picrylhydrazyl(DPPH),superoxide anion(O2-·)and hydroxyl free radicals(·OH)were tested.Protective effects of GDPs and S-GDPs on oxidative damage induced by H2O2 in erythrocytes or induced by ·OH in mice liver mitochondria were also determined.The results showed that S-GDPs had stronger antioxidative activities against DPPH(P〈0.01),O2-·(P〈0.05) and ·OH(P〈0.05) than GDPs.At the same concentrations,S-GDPs could significantly inhibit oxidative hemolysis(P〈0.01)and reduce malondialdehyde content(P〈0.05)in erythrocyte.S-GDPs could significantly reduced MDA content of mitochondria(P〈0.05),and improve the activities of succinate dehydrogenase(SDH)and ATPase(Na+-K+-ATPase and Ca2+-Mg2+-ATPase)activities(P〈0.01)in mice liver mitochondria.All the above protective effects of S-GDPs is better than GDPs,which demostrated that the antioxidant activity of polysaccharides from Gynura davaricata can be improved through sulfation.
