摘要
本文推荐估算群体药动学参数的一步法,已编成微机程序,可以应用于均匀与非均匀数据作药动学处理,当将原始数据输入微机后,经过一步计算,即可得到药动学参数的群体值。通过对23例服用茶碱治疗的哮喘病人,所获得的111对数据的运算,以及按时间分布递减到99,76,65及46对数据的运算,得到与二步法相似的结果,证明本法具有病例多,群体代表性强,个体病人取血次数少,计算快速,实用广泛等优点。
Population pharmacokinetic parameters wereestimated traditionally by the two-stage methodfrom homogenous data. The one-stage method wasrecommended for estimating population pharma-cokinetic parameters from homogenous and nonho-mogenous data. The 111 points of plasma concen-tration collected from 23 patients with asthma afteradministration of theophylline were used. The fol-lowing parameters were obtained: Ka(/h)=1.55±2.19, K(/h)=0.073±0.025, CL(1/h)=2.78±0.36. It is suggested that the one-stage method ismore rapid, convenient and simple than the two-stage method.
