摘要
目的 观察重组逆转录病毒载体转染的软骨细胞能否形成同种异体组织工程化软骨并示踪构建过程。方法 利用重组逆转录病毒载体转染的软骨细胞和组织工程材料 pluronicF 1 2 7复合物构建兔同种异体组织工程软骨 ,荧光显微镜和激光共聚焦显微镜观察其构建过程。结果 重组逆转录病毒载体转染的软骨细胞能形成组织工程软骨 ,增强型绿色荧光蛋白 (EGFP)在 3周和 6周后均得到了明显表达 ,且未影响软骨组织的形成过程。结论 重组逆转录病毒载体转染的软骨细胞能形成组织工程软骨 ,EGFP是组织工程化软骨形成过程的良好示踪剂 ,且不影响组织的形成。
Objective To determine whether the rabbit allogeneic chondrocytes, which transfected with recombinant retroviral vector PLNCX2 EGFP, could be reconstructed into the cartilage and to monitor the construction process of cartilage using allogeneic rabbit chondrocytes.Methods Rabbit allogeneic chondrocytes transfected with recombinant retroviral vector PLNCX2 EGFP in combination with tissue engineering material pluronic 127 were used to construct rabbit allogeneic tissue engineered cartilage. Fluorescent microscopy and laser scanning confocal microscopy monitored the construction process of rabbit allogeneic cartilage.Results Rabbit allogeneic chondrocytes transfected with recombinant retroviral vector PLNCX2 EGFP could construct the tissue engineered cartilage. High expression of enhanced green fluorescent protein (EGFP) was observed in engineered cartilage 3 and 6 weeks after subcutaneous injection of chondrocytes pluronic F 127 mixture. The expression of EGFP did not affect the chondrocytes' function.Conclusion Rabbit allogeneic chondrocytes transfected with recombinant retroviral vector PLNCX2 EGFP could be reconstructed into the cartilage. Expression of EGFP did not affect cartilage formation. EGFP could be used to monitor the construction process of rabbit allogeneic tissue engineered cartilage perfectly.
