摘要
目的观察远志皂苷对APP/PS1双转基因小鼠海马神经细胞线粒体功能的调控,探讨远志皂苷的神经保护作用机制。方法将APP/PS1双转基因小鼠分为模型组,远志皂苷低、中和高剂量组(18.5、37.0和74.0 mg·kg^(-1)·d^(-1)),选同月龄C57BL/6小鼠为空白对照组,采用免疫荧光检测海马神经细胞线粒体膜电位,流式细胞术检测细胞凋亡率,比色法等检测超氧化物歧化酶(SOD)、谷胱甘肽过氧化物酶(GSH-PX)活力和丙二醛(MDA)含量。结果与空白对照组相比,模型组线粒体膜电位降低,神经细胞凋亡率增高,SOD,过氧化氢酶(CAT)和(GSH-PX)活性明显降低,MDA含量明显增高;与模型组相比,远志皂苷各剂量组的线粒体膜电位升高,神经细胞凋亡率下降,SOD,CAT和GSH-PX活性明显升高,MDA含量明显降低(P<0.05)。结论远志皂苷可能通过调控线粒体功能,发挥保护海马神经元的作用。
Objective To observe the regulation of the sengenin on APP/PSI double transgenic rats' hippocampus neuronal mitochondria,explore the protection of the sengenin to the nerves.Methods APP/PSI double transgenic rats were divided into model,sengenin high,middle and low dose groups( 18. 5,37. 0 and 74. 0 mg·kg-1·d-1),the same month old C57 BL/6 rats were chosen as the control group. Hippocampus neuronal mitochondria potential was detected by immunofluorescence,cell apoptotic rate was measured by flow cytometry,SOD,GSH-PX activity and MDA content were detected by colorimetry. Results Compared with control group,mitochondrial membrane potential was decreased,the apoptotic rate of neuron cell was increased,the activity of SOD,CAT and GSH-PX were decreased significantly,the content of MDA of model group was increased significantly. Compared to model group,the mitochondrial membrane potential was increased,the apoptotic rate of neuron cell was decreased,the activity of SOD,CAT and GSH-PX were increased,the content of MDA was decreased significantly in each sengenin group( all P〈0. 05). Conclusions Sengenin could protect the function of hippocampal neurons by regulating mitochondria function.
