肾气丸对2型糖尿病阳虚证大鼠骨骼肌线粒体呼吸链复合体及ATP酶活性的影响 被引量:5

Effects of Shenqi Pill(肾气丸) on the Respiratory Chain Complexes and ATP Enzyme Activity of the Skeletal Muscle Mitochondria in Type ⅡDiabetic Rats with Yang Deficiency Syndrome

摘要 目的:观察肾气丸对2型糖尿病阳虚证大鼠骨骼肌线粒体呼吸链复合体及ATP酶活性的影响。方法:将60只SD大鼠随机分为正常组12只和造模组48只,造模组48只大鼠予以高糖高脂饲料喂养3周后,采用小剂量链脲佐菌素加氢化可的松注射造模,观察2周制备2型糖尿病(阳虚证)大鼠,随机分为模型组、肾气丸组、补阳组、罗格列酮组。给药3周后处死大鼠,采用酶联免疫分析(ELISA)法分别测定大鼠骨骼肌线粒体呼吸链复合体及ATP酶的活性。结果:模型组Na^+-K^+-ATP酶、Ca^(2+)-Mg^(2+)-ATP酶以及线粒体呼吸链复合体Ⅰ、Ⅱ、Ⅲ、Ⅳ的活性低于正常组(P<0.01);肾气丸组、罗格列酮组Na^+-K^+-ATP酶、Ca^(2+)-Mg^(2+)-ATP酶以及线粒体呼吸链复合体Ⅰ、Ⅱ、Ⅲ、Ⅳ的活性明显高于模型组(P<0.05),但组间差异无统计学意义(P>0.05);补阳组Na^+-K^+-ATP酶、Ca^(2+)-Mg^(2+)-ATP酶以及线粒体呼吸链复合体Ⅰ、Ⅱ、Ⅲ、Ⅳ的活性明显低于肾气丸组(P<0.01)。结论:肾气丸能改善2型糖尿病阳虚证大鼠骨骼肌线粒体呼吸链复合体Ⅰ、Ⅱ、Ⅲ、Ⅳ及ATP酶的活性。 [Abstractl Objective: To study the effects of Shenqi Pill on the respiratory chain complexes and ATP en- zyme activity of the skeletal muscle mitochondria in type Ⅱ diabetic rats with yang deficiency syndrnme. Methods: A total of 60 Sprague-Dawley rats were randomly divided into normal group (12 rats) and model group (48 rats), and the 48 SD rats were randomly divided into model group, Shenqi Pill group, Bu Yang group and rosiglita- zone group after used high-sugar and high-fat feed 3 weeks and small dose streptozotocin and hydrocortisone 2 weeks to induce type 2 diabetic rats with yang deficiency syndrome. The rats were killed after 3 weeks adminis- tration to measure the mitochondrial respiratory chain complex and ATP enzyme activity of the skeletal muscle by enzyme linked imnmnosorbent assay (ELISA). Results: The activity of Na+-K+-ATPase, Ca2+-Mg2+-ATPase and mitochondrial respiratory chain complexⅠ、Ⅱ、Ⅲ、Ⅳ in the model group were lower than the normal group(P〈 0.01).The activity of Na+-K+-ATPase, Ca2+-Mg2+-ATPase and mitochondrial respiratory chain complex Ⅰ、Ⅱ、Ⅲ、Ⅳ in Shenqi Pill and rosiglitazone group was significantly higher than model group (P〈0.05). However, there was no statistically significant difference between the two groups. The activity of Na+-K+-ATPase, CaZ+-Mg2+-ATPase and mitochondrial respiratory chain complex Ⅰ、Ⅱ、Ⅲ、Ⅳ in the Bu Yang group was significantly lower than Shenqi Pill group (P〈0.01).Conclusion: Shenqi Pill can improve the respiratory chain complexes Ⅰ、Ⅱ、Ⅲ、Ⅳ and ATP enzyme activity of the skeletal muscle mitochondria in type Ⅱ diabetic rats with yang deficiency syndrome.
