摘要
目的探讨体外LY294002是否通过PI3K/AKT信号通路抑制肺癌细胞自噬、增殖、侵袭和迁徙。方法采用0、10、15、20、25、30μmol·L-1 LY294002(LY294002处理组)作用于人肺癌细胞A549,以PBS替代药物作为对照组。采用CCK8实验检测LY294002处理后A549细胞增殖情况;采用Western blot检测2组A549细胞Beclin-1、LC3B(Ⅱ、Ⅰ)、PI3K、p-AKT及AKT蛋白表达水平;采用Wound healing和Transwell invasion实验检测2组A549细胞迁徙、侵袭情况。结果 LY294002能有效抑制A549细胞的增殖,且在24h呈剂量依赖性,24h IC50为19.348μmol·L-1。LY294002处理组细胞中Beclin-1、PI3K、p-AKT蛋白表达水平及LC3B(Ⅱ/Ⅰ)比率、细胞迁徙和侵袭力均显著低于对照组(P<0.05)。结论 LY294002通过抑制PI3K/AKT信号通路降低人A549细胞的自噬活性、增殖、迁徙和侵袭。
Objective To investigate whether LY294002 inhibits the autophagy,proliferation, migration and invasion of lung cancer cells through PI3K/AKT signaling pathway.Methods Hu-man lung cancer A549 cells were treated with LY294002 at concentrations of 10,15,20,25 and 30 μmol·L-1 .Control cells were treated with PBS.The proliferation of cells was detected by CCK8 assay.The expression of Beclin-1,LC3B(Ⅱ,Ⅰ),PI3K,p-AKT and AKT in A549 cells was measured by Western blot analysis.The migration and invasion of cells were determined by Wound healing and Transwell invasion assays,respectively.Results LY294002 treatment inhibi-ted the proliferation of A549 cells in a dose-dependent manner,and the 24-hour IC50 was 19.348μmol·L-1 .Compared with control group,the expression of Beclin-1,PI3K and p-AKT,ratio of LC3B Ⅱ/Ⅰ,cell migration and invasion significantly decreased in LY294002 treatment group (P 〈0.05).Conclusion LY294002 treatment suppresses the autophagy,proliferation,migration and invasion of A549 cells through inhibiting PI3K/AKT signaling pathway.
