摘要
目的:研究甘草次酸对中波紫外线(UVB)诱导人皮肤角质形成细胞(Ha Ca T)细胞光老化的保护作用及相关炎症因子影响。方法:用不同浓度的甘草次酸作用于Ha Ca T细胞,噻唑蓝(MTT)比色法测定细胞增殖率,筛选出药物最佳有效浓度;用30 m J·cm-2的UVB照射Ha Ca T细胞,建立光老化模型,用3个不同浓度的甘草次酸作用于光老化细胞,MTT比色法检测光老化Ha Ca T细胞增殖率;谷胱甘肽过氧化物酶(GSH-Px),过氧化氢酶(CAT)和乳酸脱氢酶(LDH)试剂盒分别检测甘草次酸对光老化Ha Ca T细胞中GSH-Px,CAT和LDH活性的影响;蛋白免疫印记法(Western blot)检测甘草次酸对光老化Ha Ca T细胞中白细胞介素-1β(IL-1β),IL-6和肿瘤坏死因子-α(TNF-α)蛋白表达量的影响。结果:筛选出1×10-7,1×10-6,1×10-5mol·L-1甘草次酸为最佳有效浓度。与空白组比较,UVB组细胞增殖率显著降低(P<0.01),GSH-Px,CAT活性显著降低,LDH活性显著升高(P<0.01),IL-1β,IL-6,TNF-α蛋白表达量显著升高(P<0.05,P<0.01)。与UVB组比较,1×10-7,1×10-6,1×10-5mol·L-1甘草次酸+UVB组细胞增殖率显著上升(P<0.01),GSH-Px,CAT活性显著上升,LDH活性显著降低(P<0.01),IL-1β,IL-6,TNF-α蛋白表达量显著降低(P<0.05,P<0.01)。结论:甘草次酸可通过提高GSH-Px,CAT活性,降低LDH活性,减少炎症因子IL-1β,IL-6和TNF-α的表达,从而抑制UVB诱导Ha Ca T细胞光老化。
Objective : To study the protective effect of glycyrrhizin on light aging of human keratinocytes (HaCaT) cells induced by ultraviolet B (UVB) and relevant cytokines. Method: Different concentrations of glycyrrhizin were used to treat HaCaT cells. Methyl thiazolyl tetrazolium (MTT) was used to screen the effective and safe concentration of glycyrrhizin. The light aging model was set up through the irradiation of ultraviolet radiation B (UVB) at the irradiation dose of 30 mJ·cm^-2, the activity of cells was determined by MTT method. The test kits were tested to determine the effect of glycyrrhizin on glutathione peroxidase (GSH-Px), catalase (CAT) and lactate dehydrogenase (LDH) in each group of cells. Western blot method was used to detect the effect of glycyrrhizin on interleukin-lfl (IL-1β), IL-6 and tumor necrosis factor-a (TNF-a) protein expressions. Result:The optimum effective concentrations of glycyrrhizin were 1×10^-7, 1×10^-6, 1×10^-5 mol·L^-1. Compared with the blank group, the cell proliferation rate of the UVB group significantly decreased (P 〈 0.01 ) ; the activities of GSH-Px and CAT were significantly decreased, the activity of LDH was significantly increased (P 〈 0.01 ) ; the expressions of IL-1β, IL-6 and TNF-a were increased (P 〈 0.05, P 〈 0.01 ). Compared with the UVB group, the cell proliferation rate of 1×10^-7,1×10^-6,1×10^-5mol·L^-1 glycyrrhizin + UVB group was significantly increased (P 〈 0.01 ) ; the activities of GSH-Px and CAT were significantly increased, the activity of LDH was significantly decreased (P 〈 0.01 ) ; the expressions of IL-1β, IL-6 and TNF-α were decreased (P 〈 0.05, P 〈 0.01 ). Conclusion : Glycyrrhizin can improve the activities of GSH-Px and CAT, reduce the activity of LDH, decrease the secretion of cytokines, and thus inhibit light aging of HaCaT cells induced by UVB.
