摘要
本文以^(125)Ⅰ标记血管紧张素Ⅱ([^(125)Ⅰ]AⅡ)为配基,用放射分析法测定自发性高血压大鼠(SHR)脾淋巴细胞AⅡ受体,并与血压正常的WKA大鼠进行对比观察。结果表明两种大鼠淋巴细胞在pH 7.6,22℃条件下与[^(125)Ⅰ]AⅡ发生特异性结合。并可被非标记AⅡ及AⅡ拮抗剂(Sar^1,Ile^8]AⅡ竞争抑制。经Scatchard plot分析,提示两种大鼠脾淋巴细胞上均可能存在AⅡ的高、低两种亲和力受体,且SHR淋巴细胞AⅡ受体的亲和力高于WKA大鼠。体外实验还证明AⅡ抑制大鼠淋巴细胞增殖及IL-2的产生。提示SHR的免疫功能抑制可能与淋巴细胞表达高亲和力的AⅡ受体有关。
Angiotensin Ⅱ (AⅡ) binding sites were demonstrated in rat spleen lymphocytes by using [125I]AⅡ. The binding of [125I]AⅡ to spleen lymphocytes was rapid and reversible at 22 (o'C) . It could be inhibited by unlabelled AⅡand CSar1, Ile8] AⅡ, in which IC50 was 11.0 nmo1/L and 10.0 nmol/L, respectively. There might exist two kinds of receptors with low and high affinity in rat spleen lymphocytes. The dissociation constant (KD) of high affinity sites in the spontaneously hypertensive rats (SHR) spleen lymphocytes (2.54±0.15 nmol/L) was lower than that in the WKA rat spleen lymphocytes (5.4±1.9 nmol / L). The results showed that the lymphocyte af-finity of AⅡ was higher in SHR rats than in WKA rats.AⅡ suppressed the rat lymphocyte proliferation induced by ConA and decreased their IL-2 production in vitro, especially for SHR lymphocytes. Conversely AⅡ antagonist C Sar1, Ala8] An increased the proliferation of rat lymphocytes. These results suggested that AⅡ could be responsible for the SHR T cell suppression to a certain degree.
