豚鼠触觉剥夺后桶状皮质中间神经元的实验研究

摘要 目的:观察成年豚鼠单侧触觉剥夺后其大脑两侧桶装皮质的PV和CB阳性细胞数量的差别,探讨触觉剥夺后成年豚鼠桶状皮质中间神经元改变的可能引发机制。方法:18只成年健康豚鼠随机分为2组,每组9只,制作豚鼠单侧(右侧)触觉(胡须)剥夺模型,术后常规饲养4周和8周灌注取脑,左侧大脑半球为实验侧,右侧为对照侧。用尼氏染色方法观察豚鼠桶装皮质的外形,用免疫组化法观察豚鼠大脑两侧桶装皮质PV和CB阳性神经元的变化并用统计学方法比较其数量差异。结果:成年豚鼠大脑右侧桶装皮质尼氏染色可见大量细胞聚集在一起,形成"桶"的存在,与周围细胞分界明显;左侧大脑桶状皮质细胞无细胞聚集现象。PV和CB免疫组织化学染色可见在不同时间点豚鼠两侧桶状皮质区阳性神经元数量差异明显(P<0.05),实验侧明显少于对照侧。结论:成年豚鼠单侧触觉剥夺后两侧大脑桶状皮质PV和CB阳性神经元数量明显具有差异性,可能是成年哺乳动物神经再生的表现之一。 Objective:To observe the number difference of PV and CB positive cells in adult guinea pig's two side barrel cortex after unilateral sensory deprivation,and to explore the possible mechanism of the change of middle neurons in the barrel cortex of adult guinea pigs after tactile deprivation.Methods:18 healthy adult guinea pigs were randomly divided into 2 groups and each group has 9 adult guinea pigs.The guinea Pig unilateral(right)tactile(beard)deprivation model was produced.Normal feeding was continued for 4 weeks and 8 weeks after the operation.The left hemisphere was the experimental side and the right side of the brain was for comparison.Morphology of guinea pig's barrel cortex was observed using Nissl staining method,and PV and CB positive neurons changes in guinea pig's two side brain barrel cortex was observed using the method of immunohistochemistry,and the quantity was compared using statistical method.Results:Using Nissl staining method,it could be observed that a large number of cells gathered together in adult guinea pig's right brain barrel cortex,forming apresence of 'barrel',which has an obvious demarcation to the surrounding cells;And there is no cells gathering in the left brain barrel cortex.PV and CB Immunohistochemical staining showed that there were significant positive neurons number differences in guinea pig's two side's barrel cortex at different time points(P<0.05),and the experimental side was significantly less than the control side.Conclusion:There was significant difference between the number of PV and CB positive neurons in adult guinea pig's two side barrel cortex after unilateral sensory deprivation,which may be one of the manifestations of neural regeneration in adult mammals.
