雄性激素受体抑制剂比卡鲁胺对SCID鼠骨质疏松影响的实验研究 被引量:1

The Experimental Study of the Effects of the Androgen Receptor Inhibitor Bicalutamide on Osteoporosis in SCID Rats

摘要 目的:研究雄性激素受体抑制剂(比卡鲁胺)对前列腺癌大鼠的睾酮、雌性激素的影响以及对骨质疏松的标识物的影响。方法:选取40只SCID鼠作为研究对象,正常组10只,前列腺癌模型组30只,其中前列腺癌模型组分为实验组15只,对照组15只。实验组分别给予比卡鲁胺0.2 mg/mL QD,灌胃,对照组用等量生理盐水代替。分别于给药后1、2、4周,采集实验组及相应对照组裸鼠的血液及瘤体。对睾酮、雌性激素、碱性磷酸酶、酸性磷酸酶分别用ELISA的双抗体夹心法进行检测并记录分析。结果:给药2、4周后,实验组中睾酮明显低于对照组与正常组(P<0.05),其他相关水平变化不明显(P>0.05)。给药4周后对照组和实验组雄性激素受体免疫组化平均灰度值(IOD)比较差异有统计学意义(P<0.05)。结论:比卡鲁胺对在短期对前列腺癌大鼠中的睾酮影响明显,对雌性激素无明显影响,对骨质疏松的指标影响小。 Objective:To investigate the effects of the androgen receptor inhibitor bicalutamide on testosterone and estrogen,as well as the osteoporosis markers in rats with prostate cancer.Method:Forty SCID rats were assigned to the normal group including 10 rats or the prostate cancer model group including 30 rats.The rats in the prostate cancer model group were further divided into the test group including 15 rats,and the control group including 15 rats.The test group was given bicalutamide 0.2 mg/mL QD,and rats in the control group were given normal saline following the same dosing schedule.Blood and tumor were separately collected from rats in the test group and control group at 1 week,2 weeks and 4 weeks post-treatment.Testosterone,estrogen,ALP,and ACP were separately detected by double antibody sandwich ELISA, and then recorded and analyzed.Result:Testosterone in the test group was significantly lower than in the control and normal groups(P〈0.05);there were no significant changes in any other relevant indicators(P〉0.05).After 4 weeks,compared the mean gray value of androgen receptor immunohistochemistry in the control group and the experimental group,the difference was statistically significant(P〈0.05).Conclusion:In the short term,bicalutamide has significant impact on testosterone,no significant impact on estrogen,and minor impact on the osteoporosis indicators in rats with prostate cancer.
