摘要
目的探讨鱼腥草注射液对食管癌细胞EC-109形态结构的作用。方法不同浓度(对照组、7μg/L、14μg/L、28μg/L)的鱼腥草注射液作用于食管癌细胞24h后,采用细胞计数(CCK-8)比色法检测药物对EC-109细胞的抑制作用;扫描电镜观察药物对EC-109细胞形态的变化;采用磷脂结合蛋白V/碘化丙啶(Annexin V/PI)双染法观察细胞凋亡;氨基酸自动分析仪检测药物作用前后氨基酸含量的变化。结果鱼腥草注射液对体外培养的食管癌细胞EC-109生长有明显的抑制作用,且抑制作用呈剂量依赖性;扫描电镜下,对照组细胞体积呈圆形,表面微绒毛丰富。药物作用后细胞体积缩小,表面微绒毛逐渐减少,甚至消失。对照组Annexin V和PI低染,药物作用后均高染;部分氨基酸在药物作用后有下降的趋势。结论鱼腥草注射液对食管癌细胞EC-109有明显的抑制作用,其作用机制可能是诱导细胞凋亡。
Objective To investigate the changes of Houttuynia injection on EC-109 morphology of esophageal carcinoma cells.Methods Different concentrations of Herba Houttuyniae Injection（control group,7μg/L,14μg/L,28μg/L）in esophageal carcinoma cells 24 h,the inhibitory effect of drugs on EC-109 cells was detected by CCK-8colorimetric assay.Scanning electron microscope was used to observe the changes of the morphology of EC-109 cells.Cell apoptosis was observed by Annexin V/Propidium iodide（PI）double staining method.Amino acid automatic analyzer was used to detect the changes of amino acid content before and after the drug action.Results Herba Houttuyniae injection had obvious inhibition on cultured esophageal cancer Ec-109 cell growth and the inhibition in a dose-dependent manner.Under the scanning electron microscope（SEM）,the control group cells were round,rich microvilli on the surface.After drug action,cell volume reduced,microvilli decreased,even disappeared.In the control group,V Annexin and PI were low,and the drug was highly stained.Some amino acids had a downward trend after drug action.Herba Houttuyniae injection has obvious inhibiting effect on esophageal cancer cell EC-109 and its mechanism may be the induction of apoptosis.
