摘要
目的建立一种稳定的、可操作性强的病证结合帕金森病(PD)小鼠模型。方法采用1-甲基-4-苯基-1,2,3,6-四氢吡啶(MPTP)腹腔注射法构建PD动物模型,同时设立正常对照组、复方地黄方组,从病因、症状、客观指标、药物反证等方面对PD小鼠模型进行综合判断,验证该模型的中医证候属性。结果模型组行为学、氧化应激、酪氨羟化酶(TH)mRNA及蛋白等指标与正常对照组差异显著(P<0.01)。复方地黄方组可明显改善PD小鼠的主观症状和客观指标(P<0.01)。结论 MPTP腹腔注射法制作的PD模型中医证候属性是阴虚动风证。
Objective To establish of a steady and simplify operation mice model of Parkinson's disease( PD) with combination of disease and syndrome,in order to meet the demand of study medication on treatment of PD evaluation. Methods The animal model of PD had been induced by intraperitoneal injection of MPTP.At the same time normal control,compound rehmannia prescription groups were set up to estimate PD model from the etiology,symptoms,objective index,drugs prove therapy,in order to investigate the syndrome properties of this mice model.Results There were statistically significant differences in behavior,oxidative stress,TH mRNA and protein between model and normal control groups( P〈0. 01).Compound rehmannia prescription group could significantly improve the subjective symptoms and objective indexes( P〈0.01).Conclusions The syndrome property of PD mice model induced by intraperitoneal injection of MPTP is mainly Yin Xu Dong Feng Zheng.
