摘要
丙二酸二乙基酯为起始原料,与水合肼反应得到丙二酸乙酯酰肼1,再与甲基乙亚胺酸酯盐酸盐2在Hoenig碱存在下进行缩合反应得到5-甲基-4H-1,2,4-三唑-3-乙酸酯3,最后将化合物3的酯键水解后得到目标产物5-甲基-4H-1,2,4-三唑-3-乙酸酯I,总收率40.0%。
Starting from diethyl malonate, which reacted with hydrazine hydrate to give ethyl malonate hydrazide 1, and then reacted with methyl ethylimidate hydrochloride 2 in the presence of Hoenig base through condensation to afford ethyl 5-aethy1-4H-1,2,4-triazole-3-acetate 3, and finally the ester bond of ester 3 was hydrolyzed, the targeted product 5-methy1-4H-1,2,4-triazole-3-aeetic acid I was obtained with a total yield of 40.0%.
