miR-29c在卡波西肉瘤SLK细胞增殖和侵袭过程中的作用研究

摘要 目的探讨miR-29c在卡西波内瘤(KS)细胞中的表达情况以及miR-29c在KS细胞侵袭和迁移过程中的作用及其机制。方法 qPCR检测KS细胞中miR-29c的表达情况;使用miR-29c-mimic过表达miR-29c,qPCR检测miR-29c-mimic过表达效果;Transwell侵袭实验检测miR-29c的过表达对KS细胞侵袭能力的影响;划痕实验检测miR-29c的过表达对KS细胞迁移能力的影响;CM-H2DCFDA荧光染色细胞检测KS细胞内活性氧水平。结果 miR-29c在KS SLK细胞中表达水平较低,miR-29c-mimic可以有效增加miR-29c的表达;过表达miR-29c抑制KS SLK细胞的侵袭迁移能力;过表达miR-29c后可以降低KS SLK细胞内活性氧水平。结论 miR-29c在KS细胞中表达下调,同时过表达miR-29c可以抑制KS细胞侵袭和迁移能力。 Objective To investigate the effect of miR-29 c on the invasion and migration of Kaposi's sarcoma and the related mechanism. Methods QPCR was used to detect the expression of miR-29 c in Kaposi ' s sarcoma. The overexpression of miR-29 c-mimic was detected by q PCR.Transwell invasion assay was used to detect the effect of invasion on Kaposi's sarcoma cell invasion.Scratch test invasion assay was used to detect the effect of migration on Kaposi sarcoma cell invasion.The reactive oxygen species( ROS) in Kaposi's sarcoma cells was detected by CM-H2 DCFDA staining. Results The expression of miR-29 c in Kaposi's sarcoma SLK cells was low.MiR-29 c-mimic effectively increased the expression of miR-29 c.Overexpression of miR-29 c could inhibit the invasion of SLK. Overexpression of miR-29 c reduced the level of reactive oxygen species( ROS) in Kaposi's sarcoma SLK cells.Conclusion MiR-29 c can down-regulate Kaposi's sarcoma cells,and overexpression of miR-29 c can inhibit the invasion and migration of Kaposi's sarcoma cells.
