青霉素结合蛋白测定技术的研究

摘要 当前,细菌耐药性问题日趋严重,给临床治疗带来困难,对β-内酰胺类抗生素而言,某些菌青霉素结合蛋白(PBPs)改变造成的耐药性,是β-内酰胺类抗生素的特点。 A method is described for detecting tritium-labelled penicillin-binding proteins (PBPs) of Streptococcus pneumoniae. The samples of PBPs were obtained using whole organisms treated with ~3[H]-benzyl-penicillin. Sodium lauryl sarcosinate (Sarkosyl) was found to disrupt the cytoplasmic membrane selectively and to be suitable for lysis of S. pneumoniae. This paper demonstrates that fluorography was used to record film images of ~3H in polyacrylamide gel. The distribution of radioactivity in the sample could be accurately represented by the image obtained on pre-exposed film. For detecting ~3H in polyacrylamide gels, fluorography is more sensitive than conventional autoradiography.
