摘要
目的研究miR-19a在喉鳞状细胞癌中的表达及其在体内外对喉鳞状细胞癌细胞增殖的影响。方法应用实时定量PCR检测miR-19a在喉鳞状细胞癌组织和癌旁组织中的表达差异,利用miR-19a的反义寡核苷酸慢病毒下调miR-19a的表达。在体外用MTT法观察miR-19a表达下调对喉鳞状细胞癌细胞增殖的影响。在体内,观察下调miR-19a表达对喉鳞癌裸鼠移植瘤生长的影响。结果与癌旁组织相比,miR-19a在喉鳞状细胞癌组织中显著高表达(P<0.001),下调miR-19a的表达,喉鳞癌细胞在体内外的增殖受到抑制。结论 miR-19a在喉鳞状细胞癌中高表达,下调miR-19a在体内外抑制喉鳞状细胞癌细胞的增殖。
Objective To investigate the expression of miR-19a in laryngeal squamous cell carcinoma (LSCC) and its effect on proliferation of LSCC in vivo and in vitro. Methods miR- 19a level in LSCC and adjacent non-neoplastic tissues were detected by qRT-PCR, miR-19a was knockdown by miR-19a shRNA lentivirus in LSCC cells and cell proliferation was examined by MTT/n v/fro. The growth of xenografis with miR-19a knockdown LSCC cells was analyzed. Results miR-19a level was significantly higher in LSCC than in corresponding adjacent non- neoplastic tissues (P 〈0. 001 ). Moreover, the knockdown of miR-19a significantly inhibited the proliferation of LSCC cells. The growth of LSCC xenografts was significantly suppressed by the injection of miR-19a shRNA lentivirus. Conclusion miR-19a is overexpression in LSCC. The knockdown of miR-19a could significantly inhibit the proliferation of LSCC cells in vivo and in vitro.
