ENO1在肝癌细胞株和肝细胞癌中的表达及与临床病理参数的关系 被引量:5

Expression of ENO1 in Hepatocellular Carcinoma and Its Relationship with Cinicopathological Parameters

摘要 目的探讨α-烯醇化酶(ENO1)在肝癌细胞株及肝细胞癌(HCC)组织中的表达,分析其表达与临床病理参数之间的关系。方法分别采用RT-PCR、Western blot检测32例肝细胞癌及其相应癌旁组织,L-O2细胞、HepG2细胞及HepG2.215细胞中ENO1mRNA及其蛋白的表达;用组织芯片技术检测80例肝癌组织及对应癌旁组织中ENO1的表达,并分析其与临床病理参数之间的关系。结果与癌旁肝组织相比,肝细胞癌组织中ENO1mRNA和蛋白表达均增高(P<0.05);与正常肝细胞L-O2相比,HepG2细胞和HepG2.215中ENO1mRNA和蛋白表达量均增高(P<0.05),HepG2.215细胞表达量高于HepG2.0细胞(P<0.05);组织芯片结果显示肝癌组织中ENO1蛋白表达高于癌旁组织(P<0.05),并且ENO1的表达水平与年龄、性别、甲胎蛋白(AFP)、肿瘤转移及肿瘤复发无关(P>0.05),与肿瘤大小、HBV感染有关(P<0.05)。结论 ENO1在肝癌细胞系及肝细胞癌组织中均高表达,并与肿瘤大小、HBV感染有关。 Objective To investigate the expression of ENO1 in hepatocellular carcinoma cells and hepatocellular carcinoma and its relationship with clinicopathological parameters. Methods RT-PCR,Western Blot was used to detect the expression of ENO1 mRNA and protein in 32 cases hepatocellular carcinoma and its adjacent tissues,L-O2,HepG2 and HepG2.215 cells respectively. Tissue microarray was used to detect the expression of ENO1 in 80 cases hepatocellular carcinoma tissues and its adjacent tissues,analyzed its relationship with clinicopathological parameters. Results Compared with the adjacent liver tissue,ENO1 mRNA and protein were highly expressed in hepatocellular carcinoma tissues(P〈0.05). Compared with L-O2 cell line,ENO1 mRNA and protein were highly expressed in HepG2.215 cells and HepG2.0 cells and the expression of ENO1 in HepG2.215 cells was higher than that of HepG2.0 cells(P〈0.05). And the expression level of ENO1 was not related to age,sex,AFP,tumor metastasis and tumor recurrence,but related to tumor size and HBV infection in Liver cancer tissue microarray. Conclusion ENO1 is overexpressed in hepatocellular carcinoma,associated with tumor size and HBV infection,but not related to age,sex,AFP,tumor metastasis and tumor recurrence.
