摘要
目的:探讨TGF-β1通过调节miR-99家族各miRNA对卵巢癌细胞增殖、迁移及侵袭行为的影响。方法:TGF-β1、miR-99家族各miRNA模拟物、miR-99家族各miRNA抑制物分别刺激或转染卵巢癌细胞系A2780及SKOV3。实时荧光定量PCR检测miR-99家族各miRNA表达水平。CCK-8法检测细胞增殖能力。划痕实验检测细胞迁移能力。Transwell检测细胞侵袭及迁移能力。结果:与空白对照组比较,TGF-β1处理组卵巢癌细胞中miR-99家族各miRNA表达上调(P<0.01),卵巢癌细胞增殖、迁移及侵袭能力明显增强(P<0.05);miR-99家族各miRNA模拟物转染组细胞增殖减弱(P<0.05),迁移及侵袭能力增强(P<0.05);miR-99家族各miRNA抑制物转染组细胞增殖增强(P<0.05),迁移及侵袭能力减弱(P<0.05)。结论:TGF-β1可通过调节miR-99家族表达水平促进卵巢癌细胞系A2780及SKOV3的增殖、迁移及侵袭能力,TGF-β1及miR-99家族表达水平可能作为卵巢癌恶性程度预测及预后判断的参考指标。
Objective:To investigate the effect of TGF-β1 on biological behaviors of malignance in ovarian cancer cell by regulating miR-99 family.Methods:Ovarian cancer cell line A2780 and SKOV3 were treated with TGF-β1 or transfected with miRNA mimic or inhibitor of miR-99 family.Then the expression level of miR-99 family was quantified by RT-PCR.The proliferation ability was detected by CCK-8 cell counting test.The migration capacity was examined by wound healing assay.The invasion capacity was determined by transwell assays.Results:Compared to the blank control group,there was significant upregulation of miR-99 family level after being treated with TGF-β1(P〈0.01).Compared to the blank control group,the ability of proliferation,migration and invasion was also enhanced after being treated with TGF-β1(P〈0.05);the ability of proliferation was weakened(P〈0.05),while the ability of migration and invasion was significantly strengthened(P〈0.05) after transfected with miR-99 family mimic;the ability of proliferation was enhanced(P〈0.05) while the ability of migration and invasion was significantly crippled(P〈0.05) after transfected with miR-99 family inhibitor.Conclusion:The proliferation,migration and invasion ability of ovarian cancer cell A2780 and SKOV3 were promoted by TGF-β1 via miR-99 family,which could be used in predicting the malignancy of ovarian cancer.
