摘要
采集了我国近海分布的、细鳞、尖吻、叉牙、四线列牙等5种科鱼类共25尾样品,利用PCR扩增技术对其线粒体基因COⅠ及核基因RAG2序列片段进行扩增测序,得到5种科鱼类COⅠ基因序列651bp及RAG2基因序列729bp,两基因片段均不存在插入与缺失。利用Mega 5.0计算科鱼类两基因碱基组成情况,种内与种间的遗传距离,并基于最大似然法构建了系统进化树。结果显示,系统进化树上,三线最先分化出来,位于进化树的底部,其次是细鳞,也单独形成一支,独立于其他科鱼类聚类分支之外,同属于属的3个种类并没有形成单系,与格纹岛、尖吻聚为一支;叉牙与四线列牙聚成一支,形成姐妹种,显示两者之间有较近的亲缘关系。
The 651 bp of mitochondrial DNA COⅠsequences and 726 bp of nuclear DNA RAG2 sequences from 25 individuals of 5 Teraponidae fish collected from China Sea were determined in this study.No insertion and deletion existed in the two gene fragments.Base composition and genetic distances among and within species were calculated using Mega 5.0 and the molecular phylogenetic trees were constructed using maximum likelihood methods.The results showed that in the molecular phylogenetic tree,Terapon puta was first separated and rooted at the base of the tree,the next was species T.jarbua,which also formed an independent branch and located outside the other Teraponidae clusters.Three Teraponspecies did not formed an monophyletic group,the last Teraponspecies T.theraps was clustered together with Rhynchopelates oxyrhynchus and Mesopristes cancellatus;Helotes sexlineatus and Pelates quadrilineatus were clustered tightly as sister species,indicating that there were close phylogenetic relationships between the two species.These phylogenetic results might in accordance with the view of Lee and Tsai′s report that T.theraps should be removed from genus Teraponand Helotes was combined with Pelates.
