摘要
目的观察不同浓度氯化钾(KCl)溶液刺激人脑星形胶质母细胞瘤细胞(U87)后MKK4及其下游蛋白p38MAPK的表达,初步探讨MKK4以及p38 MAPK在核酸水平的表达变化特点。方法 U87细胞采用不同浓度KCl刺激,采用反转录-聚合酶链反应(reverse transcription polymerase chain reaction,RT-PCR)方法检测MKK4及p38 MAPK mRNA表达的变化。将细胞分为对照组(0组)、不同浓度KCl组(0.001 mmol/mL,0.025 mmol/mL,0.05 mmol/mL,0.1 mmol/mL,分别标记为0.001、0.025、0.05、0.1组)。结果 RT-PCR结果显示:经KCl刺激后,各组星形胶质细胞均有MKK4以及p38 MAPK mRNA的表达,其表达量均为先上升后下降,在0.05组达到高峰,此后回落。结论 KCl刺激可使人脑星形胶质母细胞瘤细胞中MKK4以及p38 MAPK mRNA的表达增高,其表达与KCl浓度有关。
Objective To observe the expression of MKK4 and its downstream protein p38 MAPK after stimulating human astrocytoma cells(U87)with different concentrations of KCl,and to explore the expression of MKK4 and p38MAPK at the level of nucleic acid.Methods U87 cells were stimulated with different concentrations of KCl,the expression of MKK4 and p38MAPK mRNA was detected by reverse transcription polymerase chain reaction(RT-PCR).The cells were divided into 5groups:control group(group 0),different concentrations of KCl group(0.001 mmol/mL,0.025 mmol/mL,0.05 mmol/mL,0.1 mmol/mL,labeled 0.001,0.025,0.05,0.1group).Results The results of RT-PCR showed that MKK4 and p38MAPK mRNA were expressed in astrocytes after stimulation with KCl,the expression was increase and then decrease and in the 0.05 group reached the peak,then fall.Conclusion KCl stimulation can increase the expression of MKK4 and p38 MAPK mRNA in human astrocytoma cells,and its expression is related to the concentration of KCl.
