上饶早梨嫩梢热处理嫁接苗脱毒效果分析 被引量:3

Virus-free Effect Analysis of Grafted Seedlings Using Heat-treated Young Shoots of Early Pear in Shangrao

摘要 为了建立上饶早梨一种简便快速的脱毒方法,采用嫩梢进行热处理并嫁接到无毒砧木的方法,并对其嫁接苗进行苹果褪绿叶斑病毒(ACLSV)、苹果茎痘病毒(ASPV)和苹果茎沟病毒(ASGV)的RT-PCR检测。结果表明:上饶早梨嫩梢热处理嫁接苗苹果褪绿叶斑病毒(ACLSV)、苹果茎痘病毒(ASPV)和苹果茎沟病毒(ASGV)的脱除率分别为93.3%、86.7%和80%。上饶早梨嫩梢热处理嫁接可在一定程度上清除ACLSV、ASPV和ASGV梨病毒。其中,上饶早梨"黄皮消"嫩梢热处理嫁接苗ACLSV、ASPV和ASGV的病毒脱除率分别为87.5%、75%和75%,上饶早梨"六月雪"嫩梢热处理嫁接苗ACLSV、ASPV和ASGV的病毒脱除率分别为100%、100%和85.7%。因此,上饶早梨嫩梢热处理嫁接更有利于上饶早梨"六月雪"病毒的脱除。本试验结果可为上饶早梨脱毒种质离体保存库的建立提供一定的技术支持和前期脱毒材料的准备。 In order to establish a simple and rapid virus-free method of early pear in Shangrao, its young shoots were heat-treated and grafted to the non-toxic rootstock, and the apple chlorotic leaf spot virus(ACLSV) and apple stem pitting virus(ASPV) and apple stem grooving virus(ASGV) of grafted seedlings were detected by RT-PCR.The results showed that the apple chlorotic leaf spot virus(ACLSV), apple stem pitting virus(ASPV) and apple stem grooving virus(ASGV) removal rate of grafted seedlings using heat-treated young shoots of early pear in Shangrao were 93.3%, 86.7% and 80%. The heat treatment of young shoots of early pear in Shangrao could remove ACLSV, ASPV and ASGV pear virus to a certain extent. Among them, the ASPV and ACLSV grafted ASGV virus removal rate of grafted seedlings using heat-treated young shoots of the cultivar "Huangpixiao"of early pear in Shangrao were 87.5%, 75% and 75% respectively, the ASPV and ACLSV grafted ASGV virus removal rate of grafted seedlings using heat-treated young shoots of the cultivar "Liuyuexue"of early pear in Shangrao were 100%, 100% and 85.7% respectively. Therefore, the graft using heat-treated young shoots of early pear in Shangrao was more conducive to the virus removal of the cultivar"Liuyuexue"of early pear in Shangrao.The results of this experiment could provide some technical support and preparation of virus-free materials for the establishment of virus-free germplasm conservation in vitro bank of pear in Shangrao.
