摘要 目的:观察Bcl-2及转化生长因子-β1(TGF-β1)在给予黄芪多糖(APS)的麻黄素损伤小鼠肾组织中表达的变化,探讨黄芪多糖对麻黄素致损伤小鼠肾的保护作用。方法:腹腔注射麻黄素溶液后,低、高剂量APS溶液灌胃。采用比色法测定肾组织过氧化氢酶(CAT)活性的变化,免疫组织化学和免疫印迹检测肾组织Bcl-2及TGF-β1蛋白表达的变化。结果:实验对照组小鼠肾组织CAT活性显著低于空白对照组,低、高剂量APS组肾组织CAT活性显著高于实验对照组;实验对照组小鼠肾组织Bcl-2、TGF-β1蛋白表达量显著高于空白对照组,低、高剂量APS组肾组织Bcl-2、TGF-β1显著低于实验对照组。结论:APS能提高麻黄素损伤小鼠肾组织抗氧化酶活性,降低肾组织Bcl-2、TGF-β1蛋白的表达,对麻黄素致损伤小鼠肾组织有明显的保护效应。 Objective: To observe the expression changes of Bcl-2 protein and transforming growth factor-β1 (TGF-131) protein in renal tissues of mice injured by ephedrine, and to explore the protection effect of Astragalus polysaccharides(APS) on the kidney of mice injured by ephedrine. Methods: Mice were injected intraperitoneally with ephedrine and were given APS (low and high concentration) by intragastric administration. The catalase (CAT) changes of renal tissues were detected by colorimetry. The expression changes of Bcl-2 protein and TGF-β1 protein in renal tissues were measured by immunohistochemistry and Western blotting. Results: The CAT activity in the renal tissues of mice in the experimental control groups were dramatically lower than the control groups. The CAT activity of the APS groups was significantly higher than experimental control groups. The expressions of Bcl-2 and TGF-β1 protein in renal tissues of mice in the experimental control groups were higher than the control groups. The expressions of Bcl-2 and TGF-β1 protein in mice renal tissue of the APS groups were lower than the experimental control groups. Conclusion: APS can effectively enhance the antioxidant enzyme activity of mice renal tissues injured by ephedrine and reduce the expression of Bcl-2 and TGF-β1 protein in the ephedrine injured kidney. APS can effectively alleviate the damage of renal tissues caused by ephedrine.
