右美托咪啶对大鼠肝脏缺血再灌注后肺损伤的影响研究

摘要 目的探讨右美托咪啶对大鼠肝脏缺血再灌注后肺损伤的影响及可能作用机制。方法 45只SD大鼠随机均分为3组,假手术组、对照组和右美托咪定干预组。肝脏缺血再灌注后4h后,检测各组大鼠血清丙氨酸转氨酶(ALT)、天冬氨酸转氨酶(AST)水平,测定肺组织湿/干重比(W/D),ELISA法检测肺泡灌洗液内肿瘤坏死因子(TNF-α)水平,WB法检测肺组织内髓过氧化物酶(MPO)活性和超氧化物歧化酶(SOD)活性。结果与假手术组相比,对照组和右美托咪啶干预组ALT和AST水平显著升高(P<0.05),右美托咪啶干预组较对照组低。与假手术组相比,对照组和右美托咪啶干预组W/D比值显著降低(P<0.05),右美托咪啶干预组较对照组低(P<0.05)。与假手术组相比,对照组和右美托咪啶干预组TNF-α、MPO和SOD水平均显著升高(P<0.05);右美托咪啶干预组T较对照组低(P<0.05)。与假手术组相比,对照组和右美托咪啶干预组SOD水平均显著降低(P<0.05),右美托咪啶干预组较对照组高(P<0.05)。结论右美托咪啶对大鼠肝脏缺血再灌注后肺损伤具有保护作用,可能与其抗氧化、抗炎作用有关。 Objective To investigate the effect of dexmedetomidine on hepatic ischemia-reperfusion induced lung injury in rats. Methods 45 SD rats were randomly divided into 3 groups,including the sham operation group,the control group and the dexmedetomidine intervention group. 4 hours after hepatic ischemia reperfusion,their serum ALT,AST,and W/D were determined. The tumor necrosis factor( TNF alpha) in alveolar lavage fluid was detected by ELISA,and myeloperoxidase( MPO) activity and superoxide dismutase( SOD) activity in lung tissue were detected by specific kits. Results The result showed that the levels of ALT and AST increased more significantly in the control group and the dexmedetomidine intervention group were than in the sham group( P < 0. 05). The levels of ALT and AST in the dexmedetomidine intervention group was lower than in the control group( P < 0. 05). Compared with the sham operation group,W/D ratio decreased significantly in the control group and the dexmedetomidine intervention group( P < 0. 05),and it was lower in the intervention group than in the control group( P < 0. 05). Compared with the sham operation group,TNF alpha and MPO levels in the control group and the dexmedetomidine intervention group were significantly higher( P < 0. 05),and TNF alpha and MPO in the dexmedetomidine intervention group was lower than those in the control group( P < 0. 05),while SOD was lower in the control group( P < 0. 05).Conclusion Dexmedetomidine intervention has a protective effect on liver ischemia reperfusion lung injury and it might be related with its antioxidant and anti-inflammatory effect.
