摘要
目的:研究高迁移率族蛋白-1(1HMGB1)预处理的骨髓间充质干细胞(BMSCs)移植治疗大鼠心肌梗死的有效性及其机制。方法:建立急性心肌梗死(AMI)大鼠模型,体外传代培养大鼠BMSCs,以25.0μg/L HMGB1预处理细胞后48h移植到大鼠心脏梗死区域。实验分为假手术组、模型对照组、BMSCs组、HMGB1+BMSCs组。心电图检测确定心肌梗死模型的建立。细胞移植28d后进行以下检测:HE、Masson染色检测梗死心脏形态学变化;TTC染色测定心肌梗死面积;超声心动图测定心脏功能;免疫组织化学染色检测心肌梗死交界区血小板内皮细胞黏附分子-1(CD31)的表达;Western-blot检测梗死心肌组织血管内皮生长因子(VEGF)的表达。结果:心电图结果表明大鼠心肌梗死模型成功建立。与模型对照组相比,细胞移植组能有效减轻大鼠心肌纤维化程度及梗死面积,以HMGB1+BMSCs组更为明显(P<0.05)。单纯BMSCs以及HMGB1预处理BMSCs移植均能增加心肌梗死区CD31、VEGF的表达,有效改善梗死心脏功能,HMGB1预处理作用更显著(P<0.05)。结论:HMGB1预处理BMSCs较单纯的BMSCs移植更能有效减轻心肌梗死程度,改善心脏功能,并且该效应可能与HMGB1促进BMSCs的旁分泌有关。
Objective:To investigate the role of bone marrow mesenchymal stem cells(BMSCs)pretreated with high mobility group box 1(HMGB1)on myocardial infarction.Method:Rat models of acute myocardial infarction(AMI)were established.BMSCs were pretreated with 25.0μg/L of HMGB1 for 48h.The rats were divided into sham operation group,AMI model group,BMSCs transplantation group,HMGB1 preconditioning BMSCs transplantation group.Electrocardiograph was detected to determine the success of myocardial infarction models.28 days after cell transplantation,infarction area and morphological changes were detected by TTC,HE and Masson staining.Cardiac function was measured by echocardiography.Platelet-endothelial cell adhesion molecule(CD31)and vascular endothelial growth factor(VEGF)expression were determined by immunehi stochemical and Western blot.Result:The rat models of AMI were established successfully.Compared with the model AMI group,cell transplantation groups effectively reduced the degree of myocardial fibrosis and infarct size,improved the cardiac function and enhanced the expression of CD31 and VEGF.These effects above were more obvious in HMGB1 preconditioning BMSCs transplantation group(all P0.05).Conclusion:HMGB1preconditioning BMSCs transplantation is more effective than BMSCs alone in reducing myocardial infarction and improving cardiac function.The effect may be related to HMGB1 promoting BMSCs paracrine.
