白头翁皂苷D对人乳腺癌MCF-7细胞中Wnt/β-链蛋白信号通路的影响 被引量:17

摘要 目的:研究白头翁皂苷D对人乳腺癌MCF-7细胞的增殖抑制效应及其机制。方法:不同浓度的白头翁皂苷D作用于人乳腺癌MCF-7细胞48 h后,CCK-8法检测细胞生存抑制率;Hoechst法检测细胞核凋亡变化;蛋白质印迹法检测MCF-7细胞内Wnt/β-链蛋白信号蛋白及下游组件蛋白Cyclin D1和c-Myc的表达情况。结果:CCK-8法检测显示白头翁皂苷D可以抑制MCF-7细胞的增殖能力,细胞的生存抑制率呈药物浓度依赖性;Hoechst染色可见部分细胞核的形态发生改变,产生细胞核碎片,核发白等变化;与对照组比较,随着白头翁皂苷D浓度的增加,MCF-7细胞中β-链蛋白的蛋白表达量呈减少趋势;50 nmol·ml^(-1)的白头翁皂苷D诱导组细胞中Cyclin D1及c-Myc的蛋白表达量也明显减少(P<0.05)。结论:白头翁皂苷D可有效抑制人乳腺癌MCF-7细胞的增殖,其机制可能与抑制Wnt/β-链蛋白信号通路的激活,并抑制其下游蛋白Cyclin D1和c-Myc的表达有关。 Objective: To investigate the proliferation inhibition effect of pulchinenoside D on human breast carcinoma cell lines MCF-7 in vitro and its mechanism.Methods: The proliferation inhibition rate of cells was detected by CCK-8 assay after the MCF-7 cells were treated with different concentration of pulchinenoside D for 48 h,and the pathological changes of cell nucleus were determined by Hoechst assay.The protein expression of β-catenin,CyclinD1 and c-Myc was examined by Western blot analysis.Results: CCK-8 assay showed that the growth of MCF-7 cells was inhibited by pulchinenoside D in a concentration-dependent manner.Hoechst staining indicated that there were some morphological changes of the cells, such as occurrence of nuclear debris and pale.The expression of β-catenin, CyclinD1 and c-Myc was obviously decreased compared with that of the control group(P&lt;0.05).Conclusion: The growth of MCF-7 cells is significantly inhibited by pulchinenoside D, which may be involved in Wnt/β-catenin signaling pathway.
