益肾健脾方对少弱精子症小鼠模型精液质量和精子线粒体结构及功能的影响 被引量:3

Impacts of Yishen Jianpi Formula on the Semen Quality and the Structure and Function of Sperm Mitochondria in Oligoasthenotspermia Model Mice

摘要 目的观察益肾健脾方对雷公藤多苷(GTW)诱导少弱精子症小鼠模型精液质量、精子线粒体超微结构和膜电位(MMP)水平的影响,探讨益肾健脾方治疗少弱精子症可能的作用机制。方法60只BALB/c小鼠随机分为正常组、模型组、阳性对照组(东维力口服液)、中药组(益肾健脾方低、高剂量组)。除正常组外,其余各组以GTW诱导少弱精子症小鼠模型。造模后,阳性对照组给予东维力口服液灌胃,益脾健肾方低、高剂量组分别给予不同剂量的益脾健肾方灌胃,正常组和模型组给予蒸馏水灌胃。治疗结束后,应用伟力彩色精子质量检测系统检测小鼠精液质量,应用透射电镜观察精子线粒体超微结构,应用荧光染色、流式细胞技术检测精子MMP正常精子百分率(JC-1,%)。结果阳性对照组、中药组A级精子活力、精子总活力高于模型组(P<0.05);阳性对照组及中药高剂量组低于中药低剂量组(P<0.05)。阳性对照组、中药组精总数低于模型组(P<0.05);阳性对照组、中药低、高剂量组之间相比无统计学意义(P>0.05)。模型组精子MMP低于正常组(P<0.05);中药高剂量组高于中药低剂量组及阳性对照组(P<0.05);中药低剂量组高于阳性对照组(P<0.05)。结论益肾健脾方可能通过改善精子线粒体结构、提高精子MMP从而改善精液质量达到治疗少弱精子症的目的。 Objective To observe the impacts of yishen j ianpi formula on the semen quality,the mi- tochondrial ultrastructure and mitochondrial membrane potential (MMP) in the model mice of oligoasthe- notspermia induced by tripterygium glycosides (GTW) and explore the potential effect mechanism of the formula on the disease. Methods The model mice of oligoasthenotsperrnia induced by GTW were randomized into a normal group ,a model group ,a positive control group( dongweili oral liquid), Chinese herbal medicine groups( low- dose and high -dose groups of yisheu jianpi formula). The model mice received the intervention with dongweili oral liquid and Chinese herbal medicine. Weili color sperm tester was adopted to detect the semen quality in the mice, the transmission electron microscope was to observe the mitochondrial ultrastructure, the fluorescence staining and flow cytometry to detect the normal sperm percentage of MMP(JC - 1 + % ). Results The A - grade sperm motility and the total sperm motility in the positive control group and Chinese herbal medicine groups were higher than those in the model group(P 〈0.05). The results in the positive control group and the high - dose group of Chinese herbal medicine were lower than the low - dose group of Chinese herbal medicine( P 〈 0.05 ). The difference was not significant statistically in comparison of the high- dose group of Chinese herbal medicine and the positive control group( P 〉 0.05 ). The total sperm count in the positive control group and the Chinese herbal medicine groups was lower than that in the model group(P 〈 0.05 ). The difference was not significant statistically in comparison among the positive control group, the low - dose group and the high - dose group of Chinese herbal medicine ( P 〉 0.05 ). In the model group, the mitochondria were swollen apparently, the fibers were loose and the axial filament broken. After intervention,the mitochondrial sheath structure was intact and the swelling was alleviated significantly, the fi- bers and axial filament were getting neat and complete. The sperm MMP was lower than that in the normal group(P 〈0.05). After intervention, MMP in the high- dose group of Chinese herbal medicine was higher than that in the low- dose group and the positive control group( P 〈 0.05 ) ;and that in the low- dose group was higher than that in the positive control group(P 〈 0.05). Conclusion Yishen jianpi formula improves mitochondrial structure and MMP of sperms so as to increase the semen quality and treat oligoasthenotspermia.
